HMGB1和MMP-2、MMP-9在喉癌組織中的表達(dá)及其與預(yù)后的關(guān)系

【摘要】:目的探討喉癌組織中高遷移率族蛋白B1(HMGB1)和基質(zhì)金屬蛋白酶-2(MMP-2)、基質(zhì)金屬蛋白酶-9(MMP-9)的表達(dá)及其與臨床病理特征和預(yù)后的關(guān)系。 方法應(yīng)用免疫組織化學(xué)EnVision二步法檢測(cè)61例喉癌組織中HMGB1和MMP-2、MMP-9蛋白表達(dá);實(shí)時(shí)熒光定量PCR法檢測(cè)30例喉癌組織中HMGB1和MMP-2、MMP-9mRNA表達(dá)。 結(jié)果HMGB1和MMP-2、MMP-9蛋白在喉癌組織中的陽(yáng)性表達(dá)率明顯高于癌旁正常組織(χ2值分別44.934和49.923、36.054,P值均0.01);HMGB1和MMP-2、MMP-9mRNA在喉癌組織中的相對(duì)表達(dá)水平明顯高于癌旁正常組織(t值分別為5.940和7.005、7.664,P值均0.01)。HMGB1和MMP-2、MMP-9蛋白表達(dá)均與喉癌T分期、臨床分期和淋巴結(jié)轉(zhuǎn)移有關(guān)(P值均0.05);HMGB1和MMP-9蛋白在喉癌中的表達(dá)呈正相關(guān)(r=0.381,P0.01),HMGB1和MMP-2蛋白在喉癌組織中的表達(dá)無(wú)相關(guān)性(r=0.094,P0.05);Kaplan-Meier生存分析結(jié)果顯示,HMGB1和MMP-9在表達(dá)陽(yáng)性患者的生存率低于表達(dá)陰性患者(χ2值分別為4.974、6.418,P值均0.05);COX回歸分析顯示,HMGB1是危險(xiǎn)因子,其表達(dá)越高,患者預(yù)后越差(P0.05)。 結(jié)論HMGB1和MMP-2、MMP-9與喉癌的浸潤(rùn)、轉(zhuǎn)移有關(guān);HMGB1和MMP-9的表達(dá)有協(xié)同促進(jìn)作用;HMGB1是影響預(yù)后的獨(dú)立危險(xiǎn)因素。
[Abstract]:Objective to investigate the expression of high mobility group protein B1 (HMGB1), matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in laryngeal carcinoma and its relationship with clinicopathological features and prognosis. Methods the expression of HMGB1 and MMP-2,MMP-9 in 61 cases of laryngeal carcinoma was detected by immunohistochemical EnVision two-step method, and the expression of HMGB1 and MMP-2,MMP-9mRNA in 30 cases of laryngeal carcinoma was detected by real-time fluorescence quantitative PCR. Results the positive expression rate of HMGB1 and MMP-2,MMP-9 protein in laryngeal carcinoma was significantly higher than that in adjacent normal tissues (蠂 ~ 2 = 44.934 and P = 49.923 ~ 36.054, respectively). The relative expression levels of HMGB1 and MMP-2,MMP-9mRNA in laryngeal carcinoma tissues were significantly higher than those in adjacent normal tissues (t = 5.940 and 7.005 ~ 7.664g, respectively, P < 0.01). The expression of HMGB1 and MMP-2,MMP-9 protein was related to T stage, clinical stage and lymph node metastasis of laryngeal carcinoma (P < 0. 05). There was a positive correlation between the expression of HMGB1 and MMP-9 in laryngeal carcinoma (r = 0.381p 0.01), but no correlation between the expression of HMGB1 and MMP-2 protein in laryngeal carcinoma (P 0.05), Kaplan-Meier survival analysis showed that the survival rate of HMGB1 and MMP-9 in positive expression was lower than that in negative expression (蠂 2 = 4.9746.41818 P respectively). COX regression analysis showed that HMGB1 was a risk factor, the higher its expression, the worse the prognosis (P0.05). Conclusion HMGB1 and MMP-2,MMP-9 are associated with invasion and metastasis of laryngeal carcinoma, HMGB1 and MMP-9 expression are synergistic and HMGB1 is an independent risk factor for prognosis.
