Tau在鼻咽癌親本及紫杉醇耐藥細(xì)胞系中的表達(dá)差異

【摘要】:目的: 分析微管相關(guān)蛋白Tau在鼻咽癌親本細(xì)胞及紫杉醇耐藥細(xì)胞系中的表達(dá)差異,探討Tau在鼻咽癌紫杉醇耐藥中的作用。 方法: 以鼻咽癌親本細(xì)胞株CNE-1、CNE-2及其相對應(yīng)的紫杉醇耐藥細(xì)胞株CNE-1/taxol、CNE-2/taxol為實(shí)驗(yàn)對象,分別應(yīng)用Western Bolt及實(shí)時(shí)熒光定量PCR技術(shù)檢測Tau在這些細(xì)胞中的蛋白和mRNA水平的表達(dá)。 結(jié)果: 以Western Bolt和實(shí)時(shí)熒光定量PCR的結(jié)果反映Tau在不同細(xì)胞系中蛋白和mRNA水平的表達(dá)。Tau蛋白的表達(dá)水平以相對光密度參考值,即Tau蛋白的光密度參考值/同一樣本GAPDH的光密度參考值表示。mRNA的相對表達(dá)量用2-△Ct方法計(jì)算得出。結(jié)果得出,Tau蛋白在鼻咽癌親本及紫杉醇耐藥細(xì)胞系中均有表達(dá),其中耐藥細(xì)胞CNE-1/taxol、CNE-2/taxol分別是親本細(xì)胞CNE-1、CNE-2的3.11±0.54,1.39±0.21倍。Tau的mRNA在各個(gè)鼻咽癌細(xì)胞中也有表達(dá),耐藥細(xì)胞CNE-1/taxol、CNE-2/taxol分別是親本細(xì)胞CNE-1、CNE-2的2.48±0.33、1.67±0.15倍。 結(jié)論: Tau在鼻咽癌紫杉醇耐藥細(xì)胞系中的表達(dá)高于親本細(xì)胞系,Tau可能與紫杉醇耐藥相關(guān)。Tau可以作為紫杉醇化療敏感性及預(yù)后的個(gè)預(yù)測分子,對其進(jìn)行干預(yù)可以在一定程度上逆轉(zhuǎn)紫杉醇耐藥。
[Abstract]:Aim: to investigate the expression of microtubule-associated protein (Tau) in nasopharyngeal carcinoma (NPC) parent cells and paclitaxel resistant cell lines, and to explore the role of Tau in taxol resistance of nasopharyngeal carcinoma (NPC). Methods: the expression of Tau protein and mRNA in nasopharyngeal carcinoma (NPC) parent cell line CNE-1,CNE-2 and its taxol resistant cell line CNE-1/taxol,CNE-2/taxol were detected by Western Bolt and real-time fluorescence quantitative PCR. Results: the results of Western Bolt and real-time quantitative PCR showed that the expression level of Tau protein and mRNA in different cell lines was the reference value of relative optical density. The optical density reference value of Tau protein and the optical density reference value of GAPDH in the same sample were calculated by 2-Ct method. The results showed that CNE-1/taxol,CNE-2/taxol protein was expressed in nasopharyngeal carcinoma (NPC) parents and paclitaxel resistant cell lines. The expression of mRNA in resistant cells was 3.11 鹵0.54n1.39 鹵0.21 times of parental CNE-1,CNE-2, respectively. Tau was also expressed in various nasopharyngeal carcinoma cells. The CNE-1/taxol,CNE-2/taxol of drug resistant cells was 2.48 鹵0.33 1.67 鹵0.15 times of that of parental CNE-1,CNE-2, respectively. Conclusion: the expression of Tau in paclitaxel-resistant nasopharyngeal carcinoma cell line is higher than that in parent cell line. Tau may be a predictor of paclitaxel chemosensitivity and prognosis. Intervention can reverse paclitaxel resistance to some extent.
