【摘要】：目的:探討生長(zhǎng)分化因子15(GDF15)在慢性鼻-鼻竇炎(CRS)患者鼻黏膜中的表達(dá)及與黏蛋白5AC(MUC5AC)表達(dá)的關(guān)系。方法:選取15例伴鼻息肉的CRS患者,15例不伴鼻息肉的CRS患者和15例正常鼻腔黏膜,蘇木精-伊紅染色觀察各組鼻黏膜的組織形態(tài)學(xué)改變,采用PAS染色檢測(cè)各鼻黏膜中總黏蛋白的表達(dá)情況,免疫組織化學(xué)方法(IHC)、實(shí)時(shí)熒光定量PCR法(qRT-PCR)檢測(cè)各組鼻黏膜中GDF15和MUC5AC的表達(dá)情況。結(jié)果:在伴或不伴鼻息肉的CRS患者鼻腔黏膜中可觀察到黏膜增厚,杯狀細(xì)胞、腺體增生以及炎性細(xì)胞浸潤(rùn)。IHC結(jié)果顯示GDF15與MUC5AC主要分布于CRS鼻黏膜上皮及黏膜下的腺體內(nèi),較正常對(duì)照組明顯增多,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。qRT-PCR示GDF15和MUC5AC在CRS組織中的表達(dá)較正常組顯著上升(P0.05),而在伴或不伴鼻息肉的CRS組織之間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。伴或不伴鼻息肉的CRS中GDF15與MUC5AC的表達(dá)呈正相關(guān)(P0.05)。結(jié)論:GDF15和MUC5AC在伴或不伴鼻息肉的CRS患者中表達(dá)升高,且具有相關(guān)性,提示GDF15是CRS高分泌的一個(gè)重要因子。
[Abstract]:Objective: to investigate the expression of growth differentiation factor 15 (GDF 15) in nasal mucosa of patients with chronic rhinosinusitis (CRS) and its relationship with mucin 5ACMUC5AC. Methods: 15 cases of CRS with nasal polyps and 15 cases of CRS without nasal polyps and 15 cases of normal nasal mucosa were selected. The histomorphology of nasal mucosa in each group was observed by hematoxylin and eosin staining. The expression of total mucin in nasal mucosa was detected by PAS staining, and the expression of GDF15 and MUC5AC in nasal mucosa was detected by immunohistochemical method and real-time quantitative PCR method (qRT-PCR). Results: mucosal thickening, goblet cells, glandular hyperplasia and inflammatory cell infiltration were observed in nasal mucosa of CRS patients with or without nasal polyps. The results showed that GDF15 and MUC5AC were mainly distributed in the epithelial and submucous glands of CRS nasal mucosa. The expression of GDF15 and MUC5AC in CRS tissues was significantly higher than that in normal controls, but there was no significant difference between CRS tissues with or without nasal polyps. There was a positive correlation between the expression of GDF15 and MUC5AC in CRS with or without nasal polyps (P 0.05). Conclusion the expression of MUC5AC and GDF15 in CRS patients with or without nasal polyps was increased and correlated, suggesting that GDF15 is an important factor in the hypersecretion of CRS.
