miRNA-29在糖尿病視網(wǎng)膜病變患者血清中的表達(dá)分析研究

【摘要】:目的研究糖尿病視網(wǎng)膜病變患者血清中miRNA-29的表達(dá)水平,分析并探討miRNA-29在糖尿病視網(wǎng)膜病變發(fā)生機(jī)制中的作用。方法選取健康受檢者、糖尿病患者及糖尿病伴有視網(wǎng)膜病變患者,對所參加實(shí)驗(yàn)者靜脈取血,提取并純化血漿總RNA,并通過逆轉(zhuǎn)錄聚合酶鏈反應(yīng)等實(shí)驗(yàn)技術(shù)獲取并檢測miRNA-29的表達(dá)水平。結(jié)果糖尿病視網(wǎng)膜病變組患者和糖尿病不伴并發(fā)癥患者相比miRNA-29的表達(dá)顯著增高,差異具有統(tǒng)計(jì)學(xué)意義(P0.01),無并發(fā)癥的糖尿病患者組和健康對照組比較,miRNA-29的表達(dá)無明顯改變,差異無統(tǒng)計(jì)學(xué)意義(P0.05);糖尿病視網(wǎng)膜病變患者組和不伴并發(fā)癥患者糖尿病患者組、健康對照組相比,RAX蛋白表達(dá)水平明顯降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)結(jié)論miRNA-29在糖尿病視網(wǎng)膜病變患者血清中表達(dá)明顯上調(diào),miRNA-29潛在靶基因PKR結(jié)合蛋白X(RAX)表達(dá)增高,其可能參與糖尿病視網(wǎng)膜病變的發(fā)生機(jī)制,進(jìn)而引起糖尿病視網(wǎng)膜病變。
[Abstract]:Objective to study the expression level of serum miRNA-29 in diabetic retinopathy patients, analyze and discuss the role of miRNA-29 in the pathogenesis of diabetic retinopathy. Methods healthy subjects, patients with diabetes and diabetic retinopathy, to participate in the experiment were venous blood, extraction and purification of total plasma RNA expression level, and through the experimental technique of reverse transcriptase polymerase chain reaction for the detection of miRNA-29. And the results of diabetic retinopathy and diabetic patients without complication were compared to the expression of miRNA-29 was significantly increased, the difference was statistically significant (P0.01), diabetic group and healthy control group without complications, no obvious change in the expression of miRNA-29, the difference was not statistically significant (P0.05); group of patients with diabetic retinopathy and without complication in patients with diabetic patients, compared to healthy control group, The expression level of RAX protein decreased significantly, the difference was statistically significant (P0.05) conclusions over expression of miRNA-29 in serum of patients with diabetic retinopathy, the potential target gene of miRNA-29 PKR binding protein X (RAX) expression, which may be involved in pathogenesis of diabetic retinopathy, and cause diabetic retinopathy.
