【摘要】:目的觀察CXCL1在大鼠煙曲霉菌角膜炎早期角膜組織的表達情況,探討其在真菌性角膜炎固有免疫階段的作用。 方法健康Wistar大鼠66只,雌雄不限,隨機分為3組,正常對照組6只12眼,損傷對照組30只30眼,真菌感染組30只30眼,損傷對照組和真菌感染組動物均取右眼為實驗眼。刮除角膜中央上皮后涂以煙曲霉菌菌絲懸液并覆蓋自制角膜接觸鏡,建立角膜煙曲霉菌感染模型。建模后分別于4,8,16,24h處死大鼠,HE染色觀察角膜組織病理變化和中性粒細胞浸潤,運用RT-PCR和免疫組織化學技術檢測各組不同時間點CXCL1的表達情況。 結果建模后裂隙燈檢查見大鼠角膜化膿性潰瘍表現,刮取物真菌培養(yǎng)有煙曲霉菌菌落生長。HE染色見真菌感染組角膜炎癥改變,大量中性粒細胞浸潤。RT-PCR示接種真菌后4h角膜即出現CXCL1表達,并在24h內持續(xù)升高(P=0.000)。CXCL1在正常對照組和模型組角膜上皮各檢測時間點的表達量有顯著差異(各組比較P=0.000)。免疫組化結果示在角膜上皮,正常對照組有少量的CXCL1表達,損傷對照組表達升高,與正常對照組比較差異有統(tǒng)計學意義(P=0.000);真菌感染組CXCL1表達較損傷對照組更高,與正常對照組和損傷對照組比較差異有統(tǒng)計學意義(P=0.000)。 結論1、組織病理學觀察和真菌培養(yǎng)證實大鼠煙曲霉菌性角膜炎模型建立成功。2、大鼠煙曲霉菌性角膜炎早期主要特點為大量中性粒細胞浸潤。3、CXCL1基因轉錄和蛋白表達在煙曲霉菌角膜炎早期均顯著升高,提示CXCL1可能與中性粒細胞的浸潤相關。
[Abstract]:Objective to investigate the expression of CXCL1 in the early corneal tissue of rat aspergillus fumigatus keratitis and its role in the innate immune stage of fungal keratitis. Methods 66 healthy Wistar rats were randomly divided into 3 groups: normal control group (n = 6), injury control group (n = 30), fungal infection group (n = 30), normal control group (n = 6), injury control group (n = 30) and fungal infection group (n = 30). The right eyes of the injured control group and fungal infection group were taken as experimental eyes. The central epithelium of cornea was scraped with aspergillus fumigatus hyphae suspension and covered with self-made contact lens. The model of aspergillus fumigatus infection was established and the rats were killed at 4: 8 and 1624 h to observe the corneal histopathological changes and neutrophil infiltration. The expression of CXCL1 at different time points was detected by RT-PCR and immunohistochemistry. Results the corneal suppurative ulcer was detected by slit lamp in rats after modeling. The corneal inflammation was observed in the fungal culture group with aspergillus fumigatus colony growth and HE staining. A large number of neutrophils infiltrated. RT-PCR showed that there was CXCL1 expression in cornea 4 hours after inoculation. The expression of CXCL1 in corneal epithelium of normal control group and model group was significantly higher than that of control group in 24 h. The immunohistochemical results showed that there was a small amount of CXCL1 expression in corneal epithelium in normal control group and normal control group, and the expression of CXCL1 in normal control group was a little higher than that in normal control group. Compared with the normal control group, the expression of CXCL1 in the fungal infection group was higher than that in the injured control group, and the difference was statistically significant compared with the normal control group and the injured control group. Conclusion 1. Histopathological observation and fungal culture confirmed that the rat model of tobacco-curl fungal keratitis was established successfully. The main characteristics of rat early keratitis were neutrophil infiltration. 3CXCL1 gene transcription and protein surface. In the early stage of aspergillus fumigatus keratitis, The results suggest that CXCL1 may be associated with neutrophil infiltration.
