鼻咽癌裸鼠移植瘤組織的ADC值與HIF-1α的相關(guān)性研究

【摘要】:目的:檢測(cè)乏氧誘導(dǎo)因子(HIF-1α)在荷人鼻咽癌裸鼠移植瘤組織不同水平的表達(dá)及分析移植瘤組織MRI表觀彌散系數(shù)(ADC)值照射前后的動(dòng)態(tài)變化并探討二者的相關(guān)性。 方法:用免疫功能缺陷型的Balb/c裸鼠建立人鼻咽癌CNEII細(xì)胞移植瘤模型,待瘤組織平均最大直徑達(dá)1.0cm左右時(shí),將36只裸鼠隨機(jī)分成6組(每組6只)。對(duì)照組:不做任何處理;照射組給予6MV-X線,單次15Gy照射根據(jù)照射后的時(shí)間分為第1天組、第2天組、第3天組、第5天組、第7天組,MRI測(cè)量照射前、后移植瘤組織ADC值大小,采用MaxVision免疫組織化學(xué)方法檢測(cè)移植瘤組織照射前、后HIF-1α的表達(dá)情況;采用Real-time PCR和Western Blot方法檢測(cè)移植瘤組織照射前、后HIF-1α在mRNA和蛋白水平的表達(dá)水平。比較照射前、后的ADC值的變化情況及其與HIF-1α在不同水平表達(dá)的相關(guān)性。 結(jié)果:(1)HIF-1α在裸鼠移植瘤組織中有較高的表達(dá),陽(yáng)性率為69.4%。(2)照射后HIF-1αmRNA表達(dá)水平第2、3天較照射前升高,有顯著統(tǒng)計(jì)學(xué)意義,(P<0.01)照射后第7天較照射前降低;有顯著統(tǒng)計(jì)學(xué)意義,(P<0.01)。(3)照射后HIF-1α蛋白表達(dá)水平第2天較照射前升高,有顯著統(tǒng)計(jì)學(xué)意義,(P<0.01),照射后第5、7天較照射前降低;有顯著統(tǒng)計(jì)學(xué)意義,(P<0.05)。(4)照射后ADC值第2天較照射前降低,照射后第7天較照射前升高,有顯著統(tǒng)計(jì)學(xué)意義,(P<0.05)。(5)在整個(gè)標(biāo)本中,HIF-1αmRNA表達(dá)水平與ADC值呈負(fù)相關(guān),(rs=-0.727,P0.01);HIF-1α蛋白表達(dá)水平與ADC值呈負(fù)相關(guān)(rs=-0.618,P0.01),各對(duì)照組間無(wú)明顯相關(guān)性。 結(jié)論:荷人鼻咽癌裸鼠移植瘤組織中ADC值與HIF-1α的表達(dá)呈負(fù)相關(guān),這可能為ADC值預(yù)測(cè)鼻咽癌放療敏感性的機(jī)制之一,進(jìn)一步研究ADC值預(yù)測(cè)鼻咽癌放療敏感性的機(jī)制有待于進(jìn)一步研究。
[Abstract]:Aim: to detect the expression of hypoxia inducible factor (HIF-1 偽) in nude mice transplanted with human nasopharyngeal carcinoma (NPC) at different levels, and to analyze the dynamic changes of MRI diffusion coefficient before and after irradiation, and to explore the correlation between HIF-1 偽 and HIF-1 偽. Methods: the transplanted tumor model of human nasopharyngeal carcinoma (NPC) CNEII cells was established in nude mice with immunodeficient Balb/c. When the average maximum diameter of tumor tissue was about 1.0 cm, 36 nude mice were randomly divided into 6 groups (6 in each group). 6MV-X ray was given to the irradiation group. According to the time after irradiation, single 15Gy irradiation was divided into the first day group, the second day group, the third day group, the fifth day group, the 7th day group, the ADC value of the transplanted tumor tissue before and after irradiation were measured. MaxVision immunohistochemical method was used to detect the expression of HIF-1 偽 before and after irradiation, and Real-time PCR and Western Blot methods were used to detect the expression of HIF-1 偽 in mRNA and protein levels before and after irradiation. The change of ADC value and its correlation with the expression of HIF-1 偽 at different levels. Results the expression of HIF-1 偽 HIF-1 偽 in transplanted tumor tissues of nude mice was higher than that before irradiation. The positive rate was 69.40.0.The expression level of HIF-1 偽 mRNA increased on the 2nd day after irradiation compared with that before irradiation (P < 0.01). The expression rate of HIF-1 偽 was significantly lower on the 7th day after irradiation than that before irradiation. The expression level of HIF-1 偽 protein increased significantly on the 2nd day after irradiation (P < 0.01), and decreased on the 5th day after irradiation (P < 0.05), and the ADC value decreased on the second day after irradiation as compared with that before irradiation (P < 0.01), and the expression level of HIF-1 偽 protein decreased significantly at the second day after irradiation (P < 0.01), and the expression level of HIF-1 偽 protein was significantly lower than that before irradiation on the second day after irradiation (P < 0.01), and was significantly lower on the 5th and 7th day after irradiation than that before irradiation (P < 0.05). On the 7th day after irradiation, the expression of HIF-1 偽 mRNA was significantly higher than that before irradiation (P < 0.05, P < 0.05). There was a negative correlation between the expression of HIF-1 偽 mRNA and ADC value. There was a negative correlation between the expression of HIF-1 偽 protein and ADC value. There was no significant correlation between HIF-1 偽 expression level and ADC value. Conclusion: there is a negative correlation between ADC and HIF-1 偽 expression in nude mice transplanted with human nasopharyngeal carcinoma, which may be one of the mechanisms by which ADC value can predict the radiosensitivity of nasopharyngeal carcinoma. Further study on the mechanism of ADC value in predicting radiosensitivity of nasopharyngeal carcinoma needs further study.
