【摘要】：目的:初步驗證美登木素對鼻咽癌細胞株增殖以及侵襲遷移的抑制作用,利用公共數(shù)據(jù)庫芯片分析美登木素對細胞株增殖以及侵襲遷移的作用機制。方法:體外實驗檢測美登木素對鼻咽癌細胞株增殖、侵襲遷移的影響。生物信息學篩選差異表達基因、功能富集分析并進一步篩選出核心基因。分析上調(diào)、下調(diào)基因的蛋白網(wǎng)絡(PPI),并計算蛋白網(wǎng)絡以及功能富集分析的核心模塊。結(jié)果:美登木素顯著抑制鼻咽癌細胞的增殖以及侵襲遷移,生物信息學分析得到了美登木素可能影響鼻咽癌的291個上調(diào)差異基因,81個下調(diào)差異基因。PPI分析表明8個上調(diào)的差異表達基因具有顯著的生物學意義。結(jié)論:美登木素對鼻咽癌細胞株的增殖、侵襲以及遷移具有明顯的抑制作用,生物信息學表明CDKN1B、BMP2、HBEGF、SMAD6、ADM、VEGFA、CTGF、IL6等基因的異常表達可能在美登木素對鼻咽癌的侵襲、遷移的抑制中起重要作用。
[Abstract]:Objective: to investigate the inhibitory effect of medenin on the proliferation, invasion and migration of nasopharyngeal carcinoma cell lines. The mechanism of Madden lignin on proliferation, invasion and migration of nasopharyngeal carcinoma cell line was analyzed by common database chip. Methods: in vitro, the proliferation of nasopharyngeal carcinoma cell line was detected. Bioinformatics screening of differentially expressed genes, functional enrichment analysis and further screening of core genes. The protein network down-regulated the protein network of the gene and calculated the core modules of protein network and function enrichment analysis. Results: Meiden lignin significantly inhibited the proliferation and invasion and migration of nasopharyngeal carcinoma cells. Bioinformatics analysis showed that Madden lignin might affect 291 up-regulated differentially expressed genes in nasopharyngeal carcinoma, 81 down-regulated differentially expressed genes. PPI analysis showed that 8 up-regulated differentially expressed genes had significant biological significance. Proliferation of nasopharyngeal carcinoma cell lines induced by lignin, Invasion and migration were inhibited obviously. Bioinformatics indicated that the abnormal expression of CDKN1BMP2 / HBEGFSMAD6 / VEGFAA CTGFGF-IL6 may play an important role in the inhibition of invasion and migration of nasopharyngeal carcinoma induced by Madden lignin.
