【摘要】：目的:研究RTN4多態(tài)性rs2864052和rs6545468位點(diǎn)及其單倍型與廣西壯族人群鼻咽癌易感性相關(guān)關(guān)系。方法:運(yùn)用單堿基延伸PCR技術(shù)和DNA測(cè)序法對(duì)廣西壯族282例鼻咽癌患者(鼻咽癌組)和199例健康體檢者(對(duì)照組)的RTN4基因rs2864052和rs6545468位點(diǎn)進(jìn)行基因分型,并分析RTN4基因的單倍型頻率。結(jié)果:RTN4基因rs2864052存在AA、GA、GG三種基因型和A、G等位基因,其在鼻咽癌組與對(duì)照組中的基因型、等位基因頻率的分布差異無統(tǒng)計(jì)學(xué)意義;rs6545468位點(diǎn)存在CC、GC、GG三種基因型和C、G等位基因,其在鼻咽癌組與對(duì)照組中的基因型、等位基因頻率的分布差異亦無統(tǒng)計(jì)學(xué)意義。但單倍型分析發(fā)現(xiàn)鼻咽癌組單倍型AG頻率顯著低于對(duì)照組(P=0.004,OR=0.14,95%CI為0.31-0.68)。結(jié)論:RTN4基因多態(tài)性rs2864052和rs6545468位點(diǎn)的單倍型AG可降低廣西壯族人群個(gè)體鼻咽癌易感性。
[Abstract]:Objective: to study the association of RTN4 polymorphism rs2864052 and rs6545468 loci and their haplotypes with susceptibility to nasopharyngeal carcinoma (NPC) in Guangxi Zhuang nationality. Methods: single base extension PCR technique and DNA sequencing were used in 282 patients with nasopharyngeal carcinoma (NPC) in Guangxi Zhuang nationality. The rs2864052 and rs6545468 loci of RTN4 gene were genotyped in 199 healthy persons (control group) and cancer group (n = 199). The frequency of haplotype of RTN4 gene was analyzed. Results there were three genotypes and alleles in rs2864052 of RTN4 gene, which were genotypes in nasopharyngeal carcinoma (NPC) group and control group. There was no significant difference in allelic frequency distribution at rs6545468. There were three genotypes of CCG GCU GG and C G alleles, which were genotypes in nasopharyngeal carcinoma (NPC) group and control group (P < 0.05). But haplotype analysis showed that the AG frequency of haplotype in nasopharyngeal carcinoma group was significantly lower than that in control group. The CI was 0.31-0.68. Conclusion the haplotype AG at the rs2864052 and rs6545468 loci of the gene polymorphism can be decreased in Guangxi. The susceptibility of individual nasopharyngeal carcinoma in Zhuang nationality.
