AEG-1和LEF-1在眼附屬器MALT淋巴瘤的表達及意義

【摘要】:目的:研究AEG-1(astrocyte elevated gene-1)、LEF-1(即淋巴細胞增強因子)在眼附屬器粘膜相關(guān)組織淋巴瘤(marginal zone lymphoma of mucosa-associated lymphoid tissue type,MALT)和反應(yīng)性淋巴組織增生的表達,從蛋白和基因水平探討AEG-1和LEF-1的表達與眼附屬器MALT淋巴瘤發(fā)生發(fā)展關(guān)系,其蛋白表達與臨床病理特征的關(guān)系和兩者蛋白表達的相互關(guān)系及可能的機制。方法:搜集1995年1月到2015年1月青島大學附屬醫(yī)院眼科病理實驗室眼附屬器MALT淋巴瘤石蠟包埋標本和凍存組織,其中40例石蠟標本、8例凍存組織為實驗組,對照組為反應(yīng)性淋巴組織增生,其中石蠟標本20例、凍存組織8例。應(yīng)用免疫組織化學染色法檢測AEG-1和LEF-1蛋白水平的表達,應(yīng)用實時熒光定量PCR法檢測實驗組和對照組中AEG-1和LEF-1m RNA水平的表達,統(tǒng)計學分析采用SPSS17.0。結(jié)果:1.AEG-1的蛋白表達及m RNA含量:a.AEG-1陽性顆粒主要定位在細胞胞膜上,少量在胞漿內(nèi)(圖3)。b.眼附屬器MALT淋巴瘤中AEG-1的陽性表達率(62.5%)顯著高于反應(yīng)性淋巴組織增生(P0.05,表1)。c.AEG-1的陽性表達與眼附屬器MALT淋巴瘤的臨床分期有關(guān)(P0.05,表2),隨著臨床分期的增加,其表達增加,而與性別、年齡、腫瘤發(fā)生部位無關(guān)。d.AEG-1 m RNA在眼附屬器MALT淋巴瘤中顯著高于反應(yīng)性淋巴組織增生(P0.05,圖9)。2.LEF-1的蛋白表達及m RNA含量:a.LEF-1陽性顆粒主要定位在細胞胞核內(nèi)(圖6)。b.眼附屬器MALT淋巴瘤中LEF-1的陽性表達率(65%)顯著高于反應(yīng)性淋巴組織增生(P0.05,表3)。c.LEF-1的陽性表達與眼附屬器MALT淋巴瘤的臨床分期有關(guān)(P0.05,表4),隨著臨床分期的增加,其表達增加,而與患者性別、年齡、腫瘤發(fā)生部位無關(guān)。d.AEG-1 m RNA在眼附屬器MALT淋巴瘤中顯著高于反應(yīng)性淋巴組織增生(P0.05,圖10)。3.AEG-1和LEF-1蛋白表達與眼附屬器MALT淋巴瘤的關(guān)系:AEG-1和LEF-1在眼附屬器MALT組織中表達增高,陽性表達率分別為62.5%、65%,兩者之間存在正相關(guān)(P0.05,表5)Spearman相關(guān)系數(shù)為r=0.406。結(jié)論:AEG-1和LEF-1的表達增高可能參與眼附屬器MALT淋巴瘤的發(fā)生發(fā)展,兩者表達成正相關(guān),可能通過某種模式在眼附屬器MALT中發(fā)揮作用。兩種因子可能成為眼附屬器MALT淋巴瘤的診斷標記物,為眼附屬器淋巴瘤病理診斷提供依據(jù)。
[Abstract]:Objective: to study the expression of AEG-1(astrocyte elevated gene-1 lef 1 (Lymphocyte Enhancement Factor) in peripheral zone lymphoma of mucosa-associated lymphoid tissue type malt and reactive lymphoid tissue hyperplasia. To explore the relationship between the expression of AEG-1 and LEF-1 and the occurrence and development of ocular accessory MALT lymphoma from the protein and gene levels. The relationship between protein expression and clinicopathological features, the relationship between protein expression and the possible mechanism were analyzed. Methods: from January 1995 to January 2015, MALT lymph nodes were collected in ophthalmic pathology laboratory of Qingdao University Hospital. Paraffin embedded specimens and frozen tissue, The expression of AEG-1 and LEF-1 protein was detected by immunohistochemical staining method in 40 paraffin specimens and 8 frozen tissues, including 20 paraffin wax specimens and 8 frozen tissues. The expression of AEG-1 and LEF-1m RNA in the experimental group and the control group were detected by real-time fluorescence quantitative PCR method. SPSS 17.0. the results showed that the protein expression of 1 / 1 AEG-1 and the content of m RNA were mainly located on the cell membrane. The positive expression rate of AEG-1 in cytoplasm (Fig. 3n.b. ocular accessory MALT lymphoma) was significantly higher than that in reactive lymphoid tissue hyperplasia (P0.05. Table 1. C. AEG-1 was associated with the clinical staging of MALT lymphoma of ocular appendage, P 0.05, Table 2, with the increase of clinical stage. Its expression increases with sex, age, There was no correlation between tumor location and tumor location. D. The positive expression rate of LEF-1 in lymphoma was significantly higher than that in reactive lymphoproliferative tissue hyperplasia (P 0.05). The positive expression of LEF-1 was related to the clinical stage of ocular adnexal MALT lymphoma (P 0.05, Table 4), and with the increase of clinical stage, the positive expression of Lymphoma was significantly higher than that of reactive lymphoid tissue hyperplasia (P 0.05). Its expression increased with the patient's gender, age, The expression of .d.AEG-1 m RNA in ocular adnexal MALT lymphoma was significantly higher than that in reactive lymphoid tissue hyperplasia (P0.05). The relationship between the expression of AEG-1, AEG-1 and LEF-1 protein and the expression of LEF-1 in ocular adnexal MALT lymphoma was significantly higher than that in ocular adnexal MALT lymphoma. The positive expression rates were 62.5% and 65%, respectively. There was a positive correlation between them (P 0.05), and the correlation coefficient between them was 0.406.Conclusion the increased expression of LEF-1 and AEG-1 may be involved in the occurrence and development of ocular appendage MALT lymphoma, and there is a positive correlation between them. These two factors may be used as diagnostic markers of ocular accessory MALT lymphoma and provide basis for pathological diagnosis of ocular adnexal lymphoma.
