【摘要】：目的:檢測(cè)過(guò)敏性鼻炎合并哮喘(AR~+AS)患者血液嗜酸性粒細(xì)胞富集群中IL-18、IL-18BP和IL-18R的表達(dá)。方法:采集13例AR~+AS患者和14名健康人(正常對(duì)照組)外周靜脈血,用流式細(xì)胞術(shù)檢測(cè)靜息狀態(tài)下和過(guò)敏原(大籽蒿花粉、塵螨和梧桐花粉過(guò)敏原粗提液)刺激后嗜酸性粒細(xì)胞富集群中IL-18~+、IL-18BP~+和IL-18R~+細(xì)胞比例及平均熒光強(qiáng)度(MFI),分析其相關(guān)性。結(jié)果:與正常對(duì)照組相比,靜息狀態(tài)下AR~+AS患者嗜酸性粒細(xì)胞富集群中IL-18~+和IL-18BP~+細(xì)胞比例分別升高了5.821倍(P=0.001)和44.1%(P=0.048),IL-18~+細(xì)胞MFI增強(qiáng)了29.1%(P=0.001),IL-18BP~+細(xì)胞MFI降低了32.3%(P0.001),而IL-18R~+細(xì)胞比例及MFI無(wú)明顯變化(P0.05)。過(guò)敏原刺激對(duì)AR~+AS患者嗜酸性粒細(xì)胞富集群中IL-18、IL-18BP及IL-18R的表達(dá)無(wú)影響(P0.05)。AR~+AS患者血液嗜酸性粒細(xì)胞富集群中IL-18~+和IL-18BP~+細(xì)胞比例呈中度相關(guān)(r=0.622,P0.001)。結(jié)論:嗜酸性粒細(xì)胞源的IL-18和IL-18BP可能在AR~+AS中起重要作用。
[Abstract]:Objective: to detect the allergic rhinitis and asthma (AR~+AS) patients with blood eosinophil eosinophil accumulation in IL-18 group, the expression of IL-18BP and IL-18R. Methods: collected 13 cases of AR~+AS patients and 14 healthy people (normal control group) peripheral venous blood with resting cells was detected by flow cytometry and allergens (large Artemisia pollen, dust mites and pollen allergen extract Indus) stimulated eosinophils eosinophil accumulation in IL-18~+ group, IL-18BP~+ and IL-18R~+ cell proportion and mean fluorescence intensity (MFI), and the correlation was analyzed. Results: compared with normal control group, resting state AR~+AS patients with eosinophilic and IL-18~+ enrichment group the percentage of IL-18BP~+ cells were increased by 5.821 times (P=0.001) and 44.1% (P=0.048), IL-18~+ cell MFI was increased by 29.1% (P=0.001), IL-18BP~+ MFI cells decreased by 32.3% (P0.001), and IL-18R~+ cells than cases and no significant change in MFI (P0.05) allergen. Stimulation of the AR~+AS patients with eosinophilic enrichment group IL-18, had no effect on the expression of IL-18BP and IL-18R (P0.05) IL-18~+ from the blood of patients with.AR~+AS and IL-18BP~+ cell ratio was eosinophil enriched group (r=0.622, P0.001) moderate correlation. Conclusion: eosinophil derived IL-18 and IL-18BP may play an important role in AR~+AS.
