糖皮質(zhì)激素對(duì)大鼠鼻黏膜AR及SP表達(dá)的影響

【摘要】:目的:變應(yīng)性鼻炎(allergic rhinitis,AR)是鼻黏膜過(guò)敏性炎癥,AR在臨床上屬常見病多發(fā)病。因此,本研究探討糖皮質(zhì)激素對(duì)大鼠鼻黏膜AR及SP表達(dá)的影響,觀察鼻黏膜組織嗜酸性粒細(xì)胞及肥大細(xì)胞浸潤(rùn)。方法:本實(shí)驗(yàn)選用30只大鼠,采用隨機(jī)方法,分為糖皮質(zhì)激素噴鼻治療AR實(shí)驗(yàn)組及對(duì)照組。對(duì)照組包括2組,分別為AR對(duì)照組及空白對(duì)照組,每組10只。AR實(shí)驗(yàn)組及AR對(duì)照組采用卵清蛋白腹腔注射基礎(chǔ)致敏,加雙側(cè)鼻腔局部滴卵清蛋白激發(fā),建立大鼠AR模型,空白對(duì)照組用等量生理鹽水同法處理。末次局部激發(fā)后,觀察并記錄大鼠癥狀及行為變化。AR模型成功建立后,AR實(shí)驗(yàn)組用糖皮質(zhì)激素布地奈德鼻噴霧劑噴鼻治療3周,對(duì)照組用等量生理鹽水鼻噴霧劑噴鼻處理,末次噴鼻治療后,再次觀察并記錄大鼠癥狀及行為變化,水合氯醛麻醉處死3組大鼠,切取大鼠鼻黏膜組織,經(jīng)甲醛固定、脫水、石蠟、切片,分別施行HE染色、甲苯胺藍(lán)染色及免疫組化染色,顯微鏡下觀察鼻黏膜組織中嗜酸粒細(xì)胞、肥大細(xì)胞的浸潤(rùn)及SP的表達(dá)。結(jié)果:采用卵清蛋白腹腔注射基礎(chǔ)致敏,加鼻腔局部滴卵清蛋白激發(fā),成功建立了大鼠AR模型;AR實(shí)驗(yàn)組施行糖皮質(zhì)激素布地奈德鼻噴霧劑治療后,AR實(shí)驗(yàn)組大鼠搔鼻、噴嚏、流涕癥狀較生理鹽水鼻噴霧劑噴鼻治療的AR對(duì)照組明顯減少,而與空白對(duì)照組相似;AR對(duì)照組,僅用生理鹽水處理,大鼠則有明顯的搔鼻、噴嚏、流涕等AR癥狀表現(xiàn);經(jīng)3周治療,各組大鼠采用水合氯醛麻醉處死,取大鼠鼻黏膜組織進(jìn)行病理學(xué)檢查,AR實(shí)驗(yàn)組鼻黏膜組織中嗜酸性粒細(xì)胞及肥大細(xì)胞浸潤(rùn)明顯低于生理鹽水鼻噴霧劑噴鼻處理的AR對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05),而與空白對(duì)照組相似,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);糖皮質(zhì)激素布地奈德鼻噴霧劑噴鼻治療的AR實(shí)驗(yàn)組SP表達(dá)量低于AR對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05),而與空白對(duì)照組相似,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:1.卵清蛋白腹腔注射,加鼻腔局部滴入,可建立大鼠AR模型。2.糖皮質(zhì)激素布地奈德鼻噴霧劑可有效緩解AR過(guò)敏癥狀,減輕嗜酸性粒細(xì)胞及肥大細(xì)胞的浸潤(rùn),減少SP的釋放。
[Abstract]:Objective: allergic rhinitis ARA is a common disease of allergic inflammation of nasal mucosa. This study was to investigate the effect of glucocorticoid on AR and SP expression in nasal mucosa of rats, and to observe the infiltration of eosinophils and mast cells in nasal mucosa. Divided into glucocorticoid nasal spray treatment AR experimental group and control group, the control group includes two groups, AR control group and blank control group. Ten rats in each group were sensitized by intraperitoneal injection of ovalbumin (ovalbumin) and stimulated by local ovalbumin dropping in bilateral nasal cavity to establish AR model of rats. The blank control group was treated with the same amount of normal saline. After the last local excitation, the symptoms and behavioral changes of rats were observed and recorded. The AR model was established successfully. The AR experimental group was treated with glucocorticoid budesonide nasal spray for 3 weeks, the control group was treated with the same amount of saline nasal spray. After the last nasal spray treatment, the symptoms and behavior of the rats were observed and recorded again. Three groups of rats were anesthetized by chloral hydrate. The nasal mucosa of the rats was cut off, fixed by formaldehyde, dehydrated, paraffin, section, respectively, HE staining, toluidine blue staining and immunohistochemical staining. The infiltration of eosinophils and mast cells and the expression of SP in nasal mucosa were observed under microscope. Rat AR model was established successfully. After budesonide nasal spray treatment, the symptoms of nasal scratching, sneezing and runny in AR experimental group were significantly lower than those in AR control group treated with normal saline nasal spray. But similar to the blank control group. In the AR control group, the rats were treated with normal saline only, but the rats had obvious symptoms such as scratching nose, sneezing and running snot. After 3 weeks of treatment, the rats in each group were killed by chloral hydrate anesthesia, and the nasal mucosa of the rats were taken for pathological examination. The infiltration of eosinophilic granulocytes and mast cells in nasal mucosa in AR group was significantly lower than that in AR control group treated with normal saline nasal spray (P 0.05). But similar to the blank control group, the difference was not statistically significant (P 0.05). The SP expression in AR experimental group treated with budesonide nasal spray was lower than that in AR control group, the difference was statistically significant (P 0.05), but similar to that in blank control group. Conclusion: 1. Ovalbumin was injected intraperitoneally with nasal drip. Budesonide nasal spray of glucocorticoid can effectively relieve allergic symptoms of AR, reduce infiltration of eosinophils and mast cells, and reduce SP release.
