載脂蛋白E4神經(jīng)毒作用機制的研究:K~+通道抑制和Ca~(2+)超載

流行病學(xué)調(diào)查顯示,載脂蛋白E基因(apolipoprotein E gene,APOE)的ε4等位基因是阿爾茨海默病(Alzheimer’s disease,AD)的重要危險因素。研究進一步表明,AD患者腦內(nèi)的老年斑和神經(jīng)原纖維纏結(jié)中存在有較多的載脂蛋白E4(apolipoprotein E4,apoE4)。該蛋白在腦內(nèi)由星形膠質(zhì)細胞以及其它包括神經(jīng)元在內(nèi)的多種細胞合成和分泌,其過表達后發(fā)揮一定的神經(jīng)毒性作用。目前,生物化學(xué)、細胞生物學(xué)、以及轉(zhuǎn)基因動物研究已經(jīng)提出多種可能的機制來解釋apoE4與AD發(fā)病的關(guān)系,包括:aopE4在淀粉樣β蛋白的沉積和清除,以及老年斑形成中的調(diào)節(jié)作用:削弱機體的抗氧化防御系統(tǒng):擾亂細胞的信號轉(zhuǎn)導(dǎo):影響細胞骨架的結(jié)構(gòu)和功能;以及改變tau蛋白的磷酸化狀態(tài)和形成NFTs等等。多數(shù)人還認為,apoE4導(dǎo)致的胞內(nèi)Ca2+超載可能是上述各種機制的“最后共同通路”。然而,apoE4在腦內(nèi)引起Ca2+超載和損傷神經(jīng)元的確切機制尚不清楚,其初始作用以及后續(xù)或下游作用如何發(fā)生還有待深入研究。 他得注意的是,雖然細胞電活動的...
