目的 檢測(cè)幽門(mén)螺桿菌（Helicobacter pylori,H.pylori）標(biāo)準(zhǔn)株（Hp P12）和其毒力因子VacA對(duì)人胃黏膜上皮細(xì)胞（GES-1）DNA損傷和同源重組（homologous recombination,HR）修復(fù)的影響。方法 應(yīng)用Hp P12和其vacA基因敲除株（Hp P12 ΔvacA）以感染復(fù)數(shù)（multiplicity of infection,MOI）＝100分別感染GES-1細(xì)胞,采用依托泊苷（50 μmol/L）或絲裂霉素（0.5 μg/ml）處理2 h的GES-1細(xì)胞作為陽(yáng)性對(duì)照,Western blot和免疫熒光方法檢測(cè)DNA損傷標(biāo)記蛋白γH2AX、HR修復(fù)關(guān)鍵蛋白（Rad51、pMRE11、CtIP和pCtIP）的表達(dá)水平以及在DNA損傷位點(diǎn)的募集情況；進(jìn)一步采用Hp P12和Hp P12 ΔvacA菌株感染人胚腎HEK-293（DR-GFP）細(xì)胞系,驗(yàn)證VacA蛋白對(duì)HR修復(fù)的影響。結(jié)果 DNA損傷標(biāo)記蛋白γH2AX的表達(dá)和募集,在Hp P12感染組較未感染組上調(diào)（P<0.05）,在Hp P12 ΔvacA感染組較標(biāo)準(zhǔn)株感染組下調(diào)（P...
