人源重要功能新基因的克

  22周齡人胎肝是胎兒造血的主要場(chǎng)所,此階段是造血系統(tǒng)遷入和遷出的關(guān)鍵轉(zhuǎn)折點(diǎn)。這一獨(dú)特性使其極具研究?jī)r(jià)值;趯(duì)22周齡人胎肝cDNA進(jìn)行大規(guī)模測(cè)序的結(jié)果,采用生物信息學(xué)方法和實(shí)驗(yàn)手段(cDNA末端快速擴(kuò)增法等)獲得了13條具有重要功能線索的人源新基因的22條全長(zhǎng)cDNA序列。所有這些序列均率先在GenBank注冊(cè)。關(guān)于其中部分基因的主要研究結(jié)果如下: 人源Semaphorin 6C和Semaphorin 6D:克隆了Semaphorin基因家族的兩個(gè)新成員SEMA6C和SEMA6D,它們都屬于膜結(jié)合型的第六亞家族。這兩個(gè)基因分別由20個(gè)和19個(gè)外顯子組成,染色體定位分別為1q12-21.1和15q21.1。關(guān)于SEMA6C和SEMA6D,分別發(fā)現(xiàn)了3種和5種不同剪切形式,它們的表達(dá)與組織特異性及發(fā)育階段有關(guān)。Northern雜交結(jié)果顯示,SEMA6C主要在骨骼肌中表達(dá),SEMA6D則在腎、腦和胎盤(pán)中高表達(dá),在心肌和骨骼肌中也有一定量的表達(dá)。缺失體研究表明,其胞外結(jié)構(gòu)域(Sema結(jié)構(gòu)域和PSI結(jié)構(gòu)域)的完整性是Semaphorin蛋白進(jìn)行適當(dāng)?shù)姆g后修飾和亞細(xì)胞定位所必須的。SEM...
