VAPB缺失誘導(dǎo)自噬的機(jī)制研究

目的:自噬在神經(jīng)退行性疾病中扮演著重要的角色。本研究考察囊泡相關(guān)膜蛋白相關(guān)蛋白B(VAPB)是否參與自噬調(diào)控及其機(jī)制研究。方法:我們分別在HEK293T、HeLa和SH-SY5Y三種細(xì)胞系中,轉(zhuǎn)染了兩種VAPB干擾小RNA來(lái)沉默VAPB的表達(dá),利用蛋白免疫印跡法(Western blot)檢測(cè)細(xì)胞中LC3和Beclin 1的蛋白水平。在HEK293T細(xì)胞中過(guò)表達(dá)FLAG-VAPB,檢測(cè)細(xì)胞中的LC3和Beclin 1的蛋白水平。在HEK293中敲減VAPB,同時(shí)過(guò)表達(dá)GFP-LC3和FLAG-P62,通過(guò)免疫熒光的方法,檢測(cè)GFP-LC3和FLAG-P62的共定位情況。在HEK293T細(xì)胞中沉默VAPB的表達(dá),通過(guò)實(shí)時(shí)定量PCR(Real-time q PCR)檢測(cè)Beclin1 m RNA水平的變化。在HEK293T細(xì)胞中同時(shí)沉默VAPB和Beclin 1的表達(dá),檢測(cè)細(xì)胞中的LC3蛋白水平。在m Cherry-LC3穩(wěn)定轉(zhuǎn)染的HEK293細(xì)胞中,同時(shí)沉默VAPB和Beclin 1的表達(dá),檢測(cè)細(xì)胞中m Cherry-LC3點(diǎn)狀聚集的數(shù)量。在HeLa細(xì)胞中,敲減VAPB后過(guò)表達(dá)m Che...
