沉默SNHG12后BMSCs對(duì)腦缺血再灌注損傷的保護(hù)作用及機(jī)制研究

背景:缺血性腦卒中是由血液供應(yīng)中斷引起的,是世界范圍內(nèi)致死、致殘的主要原因。盡管隨著新的治療藥物和血管內(nèi)技術(shù)的發(fā)展,但全世界每年的缺血性腦卒中發(fā)病率和患病率仍在持續(xù)上升,缺血性腦卒中后發(fā)生的腦組織損傷仍然嚴(yán)重威脅著患者的生存和生活質(zhì)量。對(duì)于缺血性腦卒中后缺血再灌注損傷,目前尚無(wú)特效治療藥物,而干細(xì)胞移植可能成為最有潛力的治療方式。既往有研究報(bào)道,長(zhǎng)鏈非編碼RNA(LncRNA)在調(diào)節(jié)骨髓間充質(zhì)干細(xì)胞(BMSCs)的功能中起著關(guān)鍵作用,并有研究報(bào)道長(zhǎng)鏈非編碼RNA小核仁RNA宿主基因12(LncRNA SNHG12)在大鼠腦缺血模型腦組織中表達(dá)明顯異常。但鮮有報(bào)道SNHG12修飾的BMSCs在治療腦缺血后受損傷的腦組織中所發(fā)揮的作用。本研究主要探索沉默SNHG12后BMSCs是否通過(guò)激活PI3K/AKT-mTOR信號(hào)通路對(duì)腦缺血再灌注損傷發(fā)揮保護(hù)作用及其相應(yīng)機(jī)制。方法:本研究利用氧葡萄糖剝奪/再灌注(OGD/R)處理大鼠腦微血管內(nèi)皮細(xì)胞(BMECs),再用qPCR檢測(cè)BMECs中不同時(shí)間點(diǎn)的SNHG12的表達(dá)水平。并利用OGD/R處理的大鼠BMECs與BMSCs或OGD/R預(yù)處理的BMS...
