miR-34b/miR-23a靶向Keap1/ULK1調(diào)控腦缺血再灌注氧化損傷的機(jī)制研究

目的:探討miR-34b/miR-23a靶向Keap1/ULK1調(diào)控大鼠腦缺血再灌注氧化應(yīng)激反應(yīng)的分子機(jī)制。方法:(1)應(yīng)用QPCR技術(shù)檢測(cè)I/R大鼠腦梗死核心組織中miR-34b/miR-23a的表達(dá)水平;通過(guò)慢病毒技術(shù)在MCAO大鼠腦組織中過(guò)表達(dá)miR-34b/miR-23a,再灌注后Berdron評(píng)分與TTC染色檢測(cè)大鼠的神經(jīng)損傷和腦梗死體積,化學(xué)比色法測(cè)定總SOD、NO和MnSOD含量,ELISA檢測(cè)3-NT含量;(2)借助QPCR和Western blot技術(shù)檢測(cè)I/R大鼠腦梗死核心組織中Keap1/ULK1的表達(dá)情況,檢測(cè)miR-34b/miR-23a對(duì)Keap1/ULK1的表達(dá)調(diào)控,并用雙熒光素酶實(shí)驗(yàn)驗(yàn)證miR-34b/miR-23a與Keap1-3′-UTR/ULK1-3′-UTR的靶向結(jié)合位點(diǎn);(3)采用H2O2誘導(dǎo)細(xì)胞氧化損傷,分別上下調(diào)Keap1,過(guò)表達(dá)miR-34b和Keap1,過(guò)表達(dá)miR-23a和ULK1,再用化學(xué)比色法測(cè)定總SOD、NO與MnSOD的含量,ELISA檢測(cè)3-NT的含量。結(jié)果:(1)相較于假手術(shù)組,...
