蛛網(wǎng)膜下腔出血大鼠海馬區(qū)ROCK2信號通路對自噬的影響

目的研究蛛網(wǎng)膜下腔出血(Subarachnoid Hemorrhage,SAH)后大鼠海馬區(qū)ROCK2信號通路對自噬的影響。方法實驗用108只無特定病原體級雄性Sprague-Dawley大鼠(350450g),編碼后采用隨機數(shù)字表法等分假手術(shù)組(Sham組)、蛛網(wǎng)膜下腔出血組(SAH組)、SAH+ROCK2抑制劑組(Drug組)三個組。Sham組僅頸內(nèi)動脈分離不刺破血管,SAH模型采用頸內(nèi)動脈刺破法制作,Drug組在建模成功后給予ROCK2抑制劑(10mg/kg)干預(yù),各組分三個亞組(6h、24h、72h)。穿梭箱試驗來觀察大鼠各組的逃跑反應(yīng)次數(shù)以及逃跑反應(yīng)時間,對比各組大鼠學(xué)習(xí)記憶的改變;HE染色觀察各組大鼠海馬區(qū)神經(jīng)細(xì)胞形狀及數(shù)量變化;免疫組織化學(xué)染色及Western Blot實驗分析ROCK2蛋白以及Beclin-1和LC3-Ⅱ在大鼠海馬區(qū)的表達(dá)情況。結(jié)果1穿梭箱試驗:SAH組各亞組逃跑反應(yīng)次數(shù)減少且逃跑反應(yīng)時間增多同Sham組對比(P<0.05);Drug組亞組逃跑反應(yīng)次數(shù)增多且逃跑反應(yīng)時間減少對同SAH組對比(P<0.05)。2 HE染色結(jié)...
