柚皮苷對腦缺血再灌注損傷的保護(hù)作用

【摘要】:目的研究柚皮苷(Naringin,NRG)對大鼠局灶性腦缺血再灌注損傷的保護(hù)作用及星形膠質(zhì)細(xì)胞是否參與其中。方法柚皮苷連續(xù)灌胃7 d后進(jìn)行大腦中動(dòng)脈栓塞(middle cerebral artery occlusion,MCAO),缺血2 h后拔出線栓進(jìn)行再灌注。24 h后TUNEL染色觀察凋亡細(xì)胞數(shù)量,蘇木精-伊紅(hematoxyli-eosin,HE)染色觀察形態(tài)學(xué)改變,免疫熒光染色分別檢測星形膠質(zhì)細(xì)胞標(biāo)記物膠質(zhì)纖維酸性蛋白(glial fibrillary acidic protein,GFAP)和縫隙連接蛋白43(connexin43,Cx43)。結(jié)果同模型組相比,柚皮苷預(yù)干預(yù)后凋亡細(xì)胞數(shù)量顯著減少(P0.05),細(xì)胞損傷明顯減輕。免疫染色顯示柚皮苷可以減輕星形膠質(zhì)細(xì)胞的激活程度,同時(shí)Cx43的表達(dá)也有所降低(P0.05)。結(jié)論柚皮苷預(yù)干預(yù)可有效減輕腦缺血再灌注損傷,該保護(hù)作用同降低星形膠質(zhì)細(xì)胞Cx43表達(dá)密切相關(guān)。
[Abstract]:Objective To study the protective effect of naringenin (NRG) on focal cerebral ischemia-reperfusion injury in rats and whether astrocytes were involved. Methods The cerebral artery embolization (MCAO) was performed after 7 days of continuous administration of teak skin. After the ischemia for 2 h, the cells were reperfused. The number of apoptotic cells and hematoxylin-eosin (HE) were observed by TUNEL staining after 24 h, and the morphological changes were observed. The glial fibrillary acidic protein (GFAP) and the gap junction protein 43 (connexin43, Cx43) were detected by immunofluorescence staining. Results Compared with the model group, the number of apoptotic cells after pre-intervention of pomelo peel was significantly reduced (P0.05), and the cell damage was significantly reduced. Immunostaining showed that pomelo peel could reduce the degree of activation of astrocytes, and the expression of Cx43 was also decreased (P0.05). Conclusion The pre-intervention of pomelo peel can effectively reduce the cerebral ischemia-reperfusion injury, and the protective effect is closely related to the reduction of the expression of Cx43 in astrocyte.
