促紅細(xì)胞生成素對(duì)新生大鼠皮質(zhì)神經(jīng)元體外缺氧缺糖損傷的保護(hù)作用研究
[Abstract]:Aim to study the protective effect of rhEPO on (OGD) damage induced by oxygen-glucose deprivation in neonatal rat cortical neurons and the dose and time of rhEPO. Methods Cortical neurons of neonatal Wistar rats were cultured for 7 days and 11 days, and were randomly divided into three groups: normal control group, OGD injury group and rhEPO treated group. (1) the OGD model of cortical neurons was established with different doses of rhEPO (0.1, 1, 10 100 U/ml); (2) 10 U/ml rhEPO. were given at different time points (0 h, 24 h, 48 h after OGD injury). The morphological changes of the cells were observed and the survival rate of the cells was measured by MTT method. Results compared with the OGD injury group, rhEPO (0.1, 1, and 10 U/ml increased the survival rate of the OGD-injured cortical neurons (P0.05), and the protective effect of rhEPO was the strongest at the concentration of 10 U/ml (P0.05). The protective effect of rhEPO (10 U/ml) on cortical neurons was the strongest immediately after OGD injury (P0.05), and the survival rate of cortical neurons was not increased 24 h and 48 h after injury. Conclusion rhEPO has the protective effect on rat cortical neurons injured by hypoxia and glucose deficiency in vitro, the effective concentration of rhEPO is 0.1 脳 10 U / ml, and its concentration is 10 U/ml, and the protective effect of rhEPO is the strongest when the concentration is 10 U/ml, and the protective effect of rhEPO is the strongest immediately after the injury of neurons.
