運(yùn)動(dòng)訓(xùn)練對(duì)腦梗死大鼠梗死邊緣區(qū)神經(jīng)細(xì)胞自噬及凋亡的影響

【摘要】:目的:研究運(yùn)動(dòng)訓(xùn)練對(duì)腦梗死急性期大鼠神經(jīng)功能恢復(fù)及梗死邊緣區(qū)皮質(zhì)神經(jīng)細(xì)胞自噬及凋亡的影響。方法:建立大腦中動(dòng)脈閉塞再灌注模型,并分成運(yùn)動(dòng)訓(xùn)練組、對(duì)照組和假手術(shù)組。運(yùn)動(dòng)訓(xùn)練組大鼠給予運(yùn)動(dòng)訓(xùn)練;對(duì)照組與假手術(shù)組則不予以任何針對(duì)性訓(xùn)練。造模術(shù)后第3、7、14d采用改良神經(jīng)損傷程度評(píng)分(mNSS)對(duì)各組大鼠進(jìn)行功能評(píng)估,同時(shí)以尼氏染色法測(cè)量腦梗死體積,免疫熒光方法觀察梗死邊緣區(qū)皮質(zhì)自噬標(biāo)志物L(fēng)C3-Ⅱ及凋亡標(biāo)志物TUNEL的表達(dá)情況。結(jié)果:造模術(shù)后14d運(yùn)動(dòng)訓(xùn)練組大鼠mNSS及腦梗死體積均優(yōu)于對(duì)照組,差別具有顯著性意義(P0.05)。運(yùn)動(dòng)訓(xùn)練組大鼠梗死邊緣區(qū)皮質(zhì)LC3-Ⅱ陽(yáng)性細(xì)胞數(shù)在造模后14d少于對(duì)照組(P0.05)。相關(guān)性分析結(jié)果顯示LC3-Ⅱ陽(yáng)性細(xì)胞數(shù)與mNSS(r=0.901,P0.001)及梗死體積(r=0.832,P0.001)成正相關(guān)。TUNEL陽(yáng)性細(xì)胞數(shù)在造模后7、14d均少于對(duì)照組(P0.001),且44.6%的LC3-Ⅱ與TUNEL存在共定位。結(jié)論:運(yùn)動(dòng)訓(xùn)練能夠促進(jìn)腦梗死急性期大鼠神經(jīng)功能恢復(fù),其機(jī)制可能與其減少梗死邊緣區(qū)神經(jīng)細(xì)胞自噬及凋亡有關(guān)。
