【摘要】：目的：觀察絲氨酸/蘇氨酸激酶受體相關(guān)蛋白（Serine-threoninekinase receptor-associated protein，STRAP）及Smad家族成員7（Smadfamily member7，Smad7）在氯化鋰-匹魯卡品誘發(fā)癲癇大鼠腦組織中表達(dá)的時(shí)空變化，以探討STRAP及Smad7在顳葉癲癇形成中的作用及其機(jī)制。 方法：將60只健康雄性SD大鼠隨機(jī)分為六組：正常對(duì)照組（n=10）和經(jīng)氯化鋰-匹魯卡品誘發(fā)的癲癇組。后者再根據(jù)癲癇首次發(fā)作后的不同時(shí)間點(diǎn)分為五個(gè)實(shí)驗(yàn)組：24h（n=10），72h（n=10），1w（n=10），1m（n=10），2m（n=10）。應(yīng)用Western blot和免疫組化檢測(cè)STRAP及Smad7在氯化鋰-匹魯卡品誘發(fā)癲癇大鼠海馬及顳葉皮質(zhì)的表達(dá)變化。 結(jié)果：與正常對(duì)照組相比，癲癇大鼠腦組織中STRAP及Smad7的表達(dá)在24h,72h(急性期),1w(潛伏期),1m,2m(慢性期)顯著降低（P0.05）。 結(jié)論：STRAP及Smad7可能與顳葉癲癇形成有關(guān)，其機(jī)制主要是通過(guò)調(diào)控轉(zhuǎn)化生長(zhǎng)因子β（Transforming growth factor β,TGFβ）信號(hào)通路來(lái)實(shí)現(xiàn)。
[Abstract]:Aim: to observe the temporal and spatial changes of serine / threonine kinase receptor-associated protein (Serine-threoninekinase receptor-associated protein,STRAP) and Smad family member 7 (Smadfamily member7,Smad7) in the brain of epileptic rats induced by lithium chloride and pilocarpine. To investigate the role and mechanism of STRAP and Smad7 in temporal lobe epilepsy. Methods: sixty healthy male SD rats were randomly divided into six groups: normal control group (n = 10) and epileptic group induced by lithium chloride and pilocarpine. The latter group was divided into five experimental groups according to different time points after the first seizure: 24 h (nong 10), 72 h (nong 10), 1 w (n 10), 1 m (n 10), 2 m (n 10). Western blot and immunohistochemistry were used to detect the expression of STRAP and Smad7 in hippocampus and temporal cortex of epileptic rats induced by lithium-pilocarpine. Results: compared with the normal control group, the expression of STRAP and Smad7 in the brain of epileptic rats decreased significantly at 24 h after 72 h (acute phase), 1 week (latency), and 1 m m (chronic phase) (P0.05). Conclusion: STRAP and Smad7 may be related to the formation of temporal lobe epilepsy, and the mechanism is mainly through the regulation of transforming growth factor 尾 (Transforming growth factor 尾 (TGF 尾) signaling pathway.
