異檸檬酸脫氫酶1(IDH1)在腦膠質(zhì)瘤中的表達(dá)及意義

【摘要】:目的:通過(guò)對(duì)WHOII-III級(jí)膠質(zhì)瘤組織中異檸檬酸脫氫酶同工酶1(Isocitrate Dehydrogenase IDHl)的正常酶活性及酶蛋白含量的測(cè)定來(lái)探討膠質(zhì)瘤中IDH1基因突變表達(dá)的作用機(jī)制。 方法:56例人腦膠質(zhì)瘤組織標(biāo)本取自于2012年7月至2013年12月河北醫(yī)科大學(xué)第二醫(yī)院神經(jīng)外科手術(shù)標(biāo)本,病理診斷由醫(yī)院病理科按WHO腦腫瘤分類分級(jí)標(biāo)準(zhǔn)(2007年)進(jìn)行,其中星形細(xì)胞瘤(II級(jí))10例,少突膠質(zhì)細(xì)胞瘤(II級(jí))12例,少突星形細(xì)胞瘤(II級(jí))5例,,間變型星形膠質(zhì)細(xì)胞瘤(III級(jí))15例,間變形少突膠質(zhì)細(xì)胞瘤(III級(jí))14例。另外取腦外傷開(kāi)顱術(shù)中正常腦組織(額葉及顳葉)5例作為對(duì)照。所有腫瘤標(biāo)本及正常腦組織標(biāo)本均快速入液氮中-80℃凍存。采用酶聯(lián)免疫吸附測(cè)定法(enzyme-linked immuno sorbent assay ELISA)方法中的雙抗體夾心法檢測(cè)所有標(biāo)本組織IDH1正常酶活性。將以上標(biāo)本為3組:正常腦組織組、II級(jí)膠質(zhì)瘤組和III級(jí)膠質(zhì)瘤組。采用蛋白質(zhì)印跡法(Western Blot)同時(shí)測(cè)定3組標(biāo)本IDH1酶蛋白含量,得出相應(yīng)標(biāo)本代表蛋白含量的灰度值,采用SPSS13.0進(jìn)行統(tǒng)計(jì)分析。 結(jié)果: 1酶活性 采用ELISA法對(duì)實(shí)驗(yàn)標(biāo)本進(jìn)行酶活性值檢測(cè)。以正常腦組織組酶活性值范圍為標(biāo)準(zhǔn),將所有腫瘤組織酶活性值分為低活性組和高活性組,數(shù)值以均數(shù)±標(biāo)準(zhǔn)差表示,結(jié)果顯示:正常腦組織組(n=5)為80.297±3.357;低活性組(n=35)為48.755±6.837;高活性組(n=21)為132.246±14.887。將以上三組兩兩比較,均具有統(tǒng)計(jì)學(xué)意義(P0.05)。低活性組腫瘤例數(shù)為35,約占總腫瘤例數(shù)的62.5%。 2酶含量 采用Western Blot法對(duì)標(biāo)本進(jìn)行酶蛋白含量測(cè)定。數(shù)值以均數(shù)±標(biāo)準(zhǔn)差表示,正常腦組織組(n=5):0.2609±0.0653, II級(jí)膠質(zhì)瘤(n=27):0.4577±0.8321, III級(jí)膠質(zhì)瘤(n=29):0.7110±0.0774。以上三組兩兩比較,均具有統(tǒng)計(jì)學(xué)意義(P0.05)。 3酶活性與酶蛋白含量相關(guān)性分析 采用Pearson計(jì)算相關(guān)分析,II級(jí)膠質(zhì)瘤相關(guān)系數(shù)0.009;III級(jí)膠質(zhì)瘤相關(guān)系數(shù)0.201;所有II、III級(jí)腫瘤相關(guān)系數(shù)0.071,酶活性大小與酶含量多少相關(guān)性不明顯。 結(jié)論: 1與正常對(duì)照組相比,腦膠質(zhì)瘤組中IDH1正常酶活性存在明顯差異,活性低于正常的占62.5%,高于正常的占37.5%。結(jié)果與目前相關(guān)亞型膠質(zhì)瘤研究IDH1基因突變率相當(dāng),存在IDH1正常酶活性降低的膠質(zhì)瘤中可認(rèn)為含有IDH1基因突變。 2與正常對(duì)照組相比,膠質(zhì)瘤組織中IDH1酶蛋白含量明顯增高,且所測(cè)膠質(zhì)瘤中,II級(jí)腫瘤IDH1酶蛋白含量較III級(jí)腫瘤更高。酶蛋白表達(dá)與腫瘤級(jí)別相關(guān),提示腫瘤的惡性水平在一定程度上影響著IDH1酶蛋白的含量。 3膠質(zhì)瘤組織中,無(wú)論IDH1正常酶活性的高低與酶蛋白含量多少并無(wú)明顯相關(guān)聯(lián)系,提示雖然IDH1變異基因表達(dá)降低了IDH1正常酶活性,但并不能確定是IDH1酶蛋白含量的決定性因素,說(shuō)明腫瘤組織在IDH1基因表達(dá)上存在著各方面復(fù)雜的調(diào)控機(jī)制。
[Abstract]:Aim: to investigate the mechanism of IDH1 gene mutation expression in WHOII-III grade glioma by measuring the normal enzyme activity and the protein content of isocitrate dehydrogenase isozyme 1 (Isocitrate Dehydrogenase IDHl). Methods: from July 2012 to December 2013, 56 cases of human glioma were collected from neurosurgical department of the second Hospital of Hebei Medical University. The pathological diagnosis was performed by the Department of Pathology of the Hospital according to the WHO criteria for classification and grading of brain tumors (2007). There were 10 cases of astrocytoma (II grade), 12 cases of oligodendrocytoma (II grade), 5 cases of oligodendrocytoma (II grade), 15 cases of anaplastic astrocytoma (III grade) and 14 cases of anaplastic oligodendrocytoma (III grade). In addition, 5 cases of normal brain tissue (frontal lobe and temporal lobe) during craniotomy were taken as control. All tumor specimens and normal brain tissues were rapidly frozen at-80 鈩
