局灶性腦缺血再灌注后大鼠腦組織bFGF和PDGF-B mRNA表達(dá)及其與血管新生關(guān)系

【摘要】:目的探討局灶性腦缺血再灌注(I-R)大鼠腦組織中堿性成纖維細(xì)胞生長(zhǎng)因子(bFGF))和血小板源性生長(zhǎng)因子-B(PDGF-B)的表達(dá)及其與血管新生的關(guān)系。方法 SD大鼠48只,隨機(jī)分為假手術(shù)組(6只)和大腦中動(dòng)脈I-R(MCAI-R)組。建立SD大鼠MCAI-R模型。MCAI-R組缺血2h后根據(jù)再灌注時(shí)間窗的不同分為0h、6h、24h、3d、7d、14d、21d共7組。蘇木精-伊紅染色后觀察腦組織病理變化,原位雜交法檢測(cè)腦組織bFGF和PDGF-B mRNA的表達(dá),免疫組織化學(xué)法檢測(cè)腦組織CD34蛋白表達(dá)并計(jì)數(shù)微血管密度(MVD)。結(jié)果 MCAI-R的6h~21d組腦組織中bFGF mRNA表達(dá)與假手術(shù)組相比明顯升高,差異有顯著性(F=41.37,q=3.87~17.30,P0.05);而MCAI-R的24h~14d組與6h組相比,腦組織中bFGF mRNA表達(dá)也明顯升高,差異有顯著性(q=4.31~12.50,P0.05)。MCAI-R的6h~14d組PDGF-B mRNA表達(dá)與假手術(shù)組相比明顯升高,差異有顯著性(F=29.22,q=4.00~15.14,P0.05);而MCAI-R的24h~14d組腦組織中PDGF-B mRNA表達(dá)與MCAI-R的6h組相比,差異也有顯著性(q=3.29~11.14,P0.05)。MCAI-R的24h~14d組腦組織中MVD表達(dá)與假手術(shù)組相比均明顯升高,差異有顯著性(F=18.42,q=4.69~11.67,P0.05)。MCAI-R的3d~14d組腦組織中MVD與MCAI-R的6h組相比也明顯升高,差異有統(tǒng)計(jì)學(xué)意義(q=6.06~10.06,P0.05)。且腦組織中bFGF、PDGF-B mRNA表達(dá)與MVD呈明顯正相關(guān)(r=0.725、0.784,P0.01)。結(jié)論局灶性腦I-R后腦組織中bFGF與PDGFB mRNA表達(dá)升高,并一定程度上促進(jìn)新生血管形成,這對(duì)缺血性腦損傷有重要的保護(hù)作用。
[Abstract]:Objective to investigate the expression of basic fibroblast growth factor (bFGF) and platelet-derived growth factor B (PDGF-B) in focal cerebral ischemia-reperfusion (I-R) rats and their relationship with angiogenesis. Methods 48 SD rats were randomly divided into sham-operated group (n = 6) and middle cerebral artery (MCAI-R) group. The MCAI-R model of SD rats was established. The MCAI-R group was divided into 7 groups according to the time window of reperfusion 2 hours after ischemia. The pathological changes of brain tissue were observed after hematoxylin-eosin staining, the expression of bFGF and PDGF-B mRNA in brain tissue was detected by in situ hybridization, the expression of CD34 protein in brain tissue was detected by immunohistochemistry and the microvessel density (MVD).) was counted. Results the expression of bFGF mRNA in 6h~21d group of MCAI-R was significantly higher than that in sham operation group (P 0.05). The expression of bFGF mRNA in brain tissue of MCAI-R 24h~14d group was significantly higher than that of 6 h group, and the difference was significant (P 0.05). The PDGF-B mRNA expression in MCAI-R 6h~14d group was significantly higher than that in sham operation group (P 0.05), and the expression of PDGF-B mRNA in 6h~14d group was significantly higher than that in sham operation group (P < 0.05). The difference was significant (FG 29.22QD 4.00 15.14 P 0.05); There was also a significant difference in the expression of PDGF-B mRNA between the 24h~14d group of MCAI-R and the 6 h group of MCAI-R (qn3.29 11.14 P 0.05). The expression of MVD in the 24h~14d group of MCAI-R was significantly higher than that in the sham operation group (P < 0.05), and the expression of MVD in the brain tissue of the MCAI-R group was significantly higher than that of the sham operation group. The brain tissue MVD in MCAI-R group was significantly higher than that in MCAI-R group (P 0.05). There was a positive correlation between the expression of bFGF,PDGF-B mRNA and MVD in brain tissue (r = 0.725, 0.784, P0.01). Conclusion the expression of bFGF and PDGFB mRNA in the brain tissue after focal brain I-R is increased, and the angiogenesis is promoted to some extent, which plays an important role in the protection of ischemic brain injury.
