凝血酶和血紅蛋白對大鼠水通道蛋白的影響及與腦積水的相關(guān)性研究

【摘要】:目的探討凝血酶和血紅蛋白對水通道蛋白(AQP)的影響及AQP與腦積水的相關(guān)性。方法取清潔級健康雄性SD大鼠84只,采用隨機數(shù)字表法分為對照組、凝血酶組及血紅蛋白組,分別通過枕大池注射等滲鹽水(0.3 ml)、凝血酶[0.3 ml(10 U/ml)]及血紅蛋白[0.3 ml(150 mg/ml)]制作腦積水模型,按照缺失補足的方式,保持各組大鼠24只。觀察造模后1、3、7、14 d時的相對側(cè)腦室面積、AQP1和AQP4的表達(dá)及AQP與相對側(cè)腦室面積的相關(guān)性。結(jié)果 (1)相對于對照組,凝血酶組與血紅蛋白組在1、3、7、14 d均顯示腦積水,且在1 d時最明顯,分別為[(6.94±0.19)%和(6.58±0.15)%比(3.40±0.13)%、(6.06±0.12)%和(5.79±0.09)%比(3.55±0.15)%、(5.80±0.13)%和(5.58±0.08)%比(3.78±0.18)%、(5.66±0.14)%和(5.47±0.13)%比(3.52±0.18)%],差異均有統(tǒng)計學(xué)意義(P0.01)。(2)AQP1的增加主要在腦室脈絡(luò)叢上皮細(xì)胞基底膜與頂膜,AQP4的增加主要在腦室室管膜細(xì)胞。1、3、7、14 d時AQP1與AQP4的相對表達(dá)量,對照組分別為1.09±0.07和1.30±0.15、0.91±0.06和1.18±0.12、1.33±0.17和1.16±0.08、1.22±0.11和1.00±0.10,凝血酶組分別為4.40±0.14和3.69±0.11、3.88±0.11和3.17±0.07、3.55±0.07和2.86±0.13、3.36±0.07和2.70±0.07,血紅蛋白組分別為4.24±0.07和3.55±0.10、3.77±0.08和3.04±0.09、3.46±0.07和2.76±0.08、3.31±0.10和2.62±0.08;凝血酶組與血紅蛋白組在各時點的AQP1與AQP4的相對表達(dá)量均明顯高于對照組,組間差異均有統(tǒng)計學(xué)意義(均P0.01);血紅蛋白組各時點的AQP1與AQP4 mRNAs的相對表達(dá)量與凝血酶組的差異均無統(tǒng)計學(xué)意義(均P0.05);在凝血酶組和血紅蛋白組中,相對于1 d時,AQP1與AQP4的表達(dá)在3、7、14 d有明顯下降(均P0.01);相對于3 d時,AQP1在7、14 d均有明顯下降(均P0.05),AQP4在14 d時下降,差異均有統(tǒng)計學(xué)意義(均P0.05)。(3)凝血酶組各時點AQP1(r=0.983,P0.01)及AQP4(r=0.987,P0.01)相對表達(dá)量與相對側(cè)腦室面積呈正相關(guān);血紅蛋白組各時點AQP1(r=0.9 6 4,P0.0 1)及AQP4(r=0.9 6 2,P0.0 1)相對表達(dá)量與相對側(cè)腦室面積呈正相關(guān)。結(jié)論蛛網(wǎng)膜下腔注射凝血酶與血紅蛋白后可引起腦室及其周圍區(qū)域AQP1與AQP4的表達(dá)增加,凝血酶與血紅蛋白可能是蛛網(wǎng)膜下腔出血后腦積水的重要介導(dǎo)因素。
[Abstract]:Objective to investigate the effect of thrombin and hemoglobin on aquaporin (AQP) and the correlation between AQP and hydrocephalus. Methods 84 healthy male SD rats of clean grade were randomly divided into three groups: control group, thrombin group and hemoglobin group. The rats were injected with isoosmotic saline (0.3 ml),) through the cistern of the occipital. Hydrocephalus model was made by thrombin [0. 3 ml (10 U/ml)] and hemoglobin (0. 3 ml (150 mg/ml). The relative ventricular area, the expression of AQP1 and AQP4, and the correlation between AQP and the relative ventricular area were observed at 14 days after the establishment of the model. Results (1) compared with the control group, hydrocephalus was found in the thrombin group and the hemoglobin group on the 1st day (6.94 鹵0.19)% and (6.58 鹵0.15)% vs (3.40 鹵0.13)%, respectively. (6.06 鹵0.12)% and (5.79 鹵0.09)% vs (3.55 鹵0.15)%, (5.80 鹵0.13)% and (5.58 鹵0.08)% vs (3.78 鹵0.18)%, respectively. (5. 66 鹵0. 14)% vs (5. 47 鹵0. 13)% vs (3. 52 鹵0. 18)%, the difference was statistically significant (P0. 01). (2) AQP1 increased mainly in the basement membrane and parietal membrane of choroid plexus epithelial cells. The increase of AQP4 was mainly in ventricular ependymal cells. In the control group, 1.09 鹵0.07 and 1.30 鹵0.15U 0.91 鹵0.06 and 1.18 鹵0.121.33 鹵0.17 and 1.16 鹵0.08U 1.22 鹵0.11 and 1.00 鹵0.10, respectively. The thrombin group was 4.40 鹵0.14 and 3.69 鹵0.111.88 鹵0.11 and 3.17 鹵0.07 鹵0.07, 3.55 鹵0.07 and 2.86 鹵0.13, 3.36 鹵0.07 and 2.70 鹵0.07, respectively. The hemoglobin group was 4.24 鹵0.07 and 3.55 鹵0.10, 3.77 鹵0.08 and 3.04 鹵0.09, 3.46 鹵0.07 and 2.76 鹵0.08, 3.31 鹵0.10 and 2.62 鹵0.08, respectively. The relative expression of AQP1 and AQP4 in thrombin group and hemoglobin group was significantly higher than that in control group (P0.01). There was no significant difference in the relative expression of AQP1 and AQP4 mRNAs between hemoglobin group and thrombin group (P0.05). In the thrombin group and hemoglobin group, the expression of AQP1 and AQP4 decreased significantly at 714 days (all P0.01). Compared with 3 days, AQP1 decreased significantly at 7d (P0.05) and AQP4 decreased at 14d (P0.05). The difference was statistically significant (P0.05). (3), AQP1 (RP0.01) and AQP4 (RP0. 987) in thrombin group at each time point. P0.01) the relative expression was positively correlated with the area of the lateral ventricle. There was a positive correlation between the relative expression of AQP1 (r = 0.964) and AQP4 (r = 0.9062) and the area of the lateral ventricle in the hemoglobin group at each time point. Conclusion Subarachnoid injection of thrombin and hemoglobin can increase the expression of AQP1 and AQP4 in the ventricle and its surrounding regions. Thrombin and hemoglobin may be the important mediating factors of hydrocephalus after subarachnoid hemorrhage.
