跑臺(tái)訓(xùn)練對(duì)慢性腦缺血大鼠海馬區(qū)Notch1和突觸素表達(dá)的影響
[Abstract]:Aim: to study the effects of treadmill training on the expression of Notch1 and synaptophysin (SYN) in hippocampus of rats with chronic cerebral ischemia, and to explore the rehabilitative effect of treadmill training on chronic cerebral ischemia and its possible molecular mechanism. Methods: ninety male SD rats were randomly divided into 9 groups, 3 sham-operated control groups (7 days, 14 days, 28 days), 3 model groups (7 days, 14 days, 28 days) and 3 treadmill training groups (7 days, 14 days, 28 days), with 10 rats in each group. The model of chronic cerebral ischemia was established by permanent ligation of bilateral common carotid artery. The treadmill training group began to run on the third day after operation, 1 week training 6 days. The changes of Notch1 and synaptophysin expression in hippocampus of rats were analyzed by Western-blot and immunohistochemistry at each time point. Results: compared with sham-operation group, Notch1 protein content and expression in hippocampus of model group and treadmill group were significantly decreased at 28 days (P0.05), especially in model group compared with treadmill group (P0.05). The content and expression of synaptophysin (synaptophysin,SYN) in model group and treadmill training group were lower than that in sham operation group. The protein content and expression of Notch1 and SYN in the treadmill training group were significantly higher than those in the model group at 28 days (P0.05). Conclusion: treadmill training can increase the expression of Notch1 and synaptophysin in hippocampus of rats with chronic cerebral ischemia.
