TRPC3在海人酸誘導(dǎo)的癲癇發(fā)作大鼠大腦皮質(zhì)中表達(dá)上調(diào)

【摘要】:目的研究瞬時受體電位通道蛋白3(transient receptor potential canonical 3,TRPC3)在海人酸(kainic acid,KA)致癇大鼠大腦皮質(zhì)中的表達(dá)變化,探討其與癲癇發(fā)病的關(guān)系及意義。方法選取54只成年雄性SD大鼠隨機(jī)分為對照組(NS組)18只和實驗組(KA組)36只,實驗組給予側(cè)腦室注射KA 2μg/kg(7μL左右)制作KA致癇模型,對照組給予側(cè)腦室注射等量生理鹽水,觀察記錄大鼠癲癇發(fā)作的行為學(xué)表現(xiàn)。于癲癇發(fā)作后不同時間點(48h、72h)各取18只大鼠注射側(cè)皮層腦組織,對照組于注射結(jié)束后48h取18只大鼠同側(cè)皮層腦組織,用半定量RT-PCR、Western blot和免疫組織化學(xué)技術(shù)(均各6只)檢測TRPC3的mRNA和蛋白的含量。結(jié)果對照組大鼠無癲癇發(fā)作,實驗組大鼠于注射后5min左右出現(xiàn)典型癇性發(fā)作,達(dá)Ⅳ~Ⅴ級,持續(xù)數(shù)小時;實驗組TRPC3的mRNA和蛋白表達(dá)水平在注射后48h和72h均較對照組顯著增高(P0.05),但實驗組兩個時間點之間差異無統(tǒng)計學(xué)意義。結(jié)論 TRPC3在致癇大鼠大腦皮質(zhì)中的表達(dá)增加,其可能參與了癲癇的形成和發(fā)展。
[Abstract]:Objective to investigate the expression of transient receptor channel protein 3 (transient receptor potential canonical 3 (TRPC3) in the cerebral cortex of epileptic rats induced by kainic acid (kainic acid,KA) and its relationship with epilepsy. Methods Fifty-four adult male SD rats were randomly divided into control group (NS group, n = 18) and experimental group (KA group, n = 36). The experimental group was given intraventricular injection of KA 2 渭 g/kg (about 7 渭 L) to make the model of KA induced epilepsy. Rats in the control group were given the same amount of saline to observe the behavior of epileptic seizures. Eighteen rats were injected with lateral cortical brain tissue at different time points (48h / 72h) after seizure, and 18 rats' ipsilateral cortical brain tissue were taken from control group at 48h after injection. Semi-quantitative RT-PCR, was used. The contents of mRNA and protein in TRPC3 were detected by Western blot and immunohistochemistry. Results there was no epileptic seizure in the control group, and typical epileptic seizure occurred in the experimental group after injection of 5min, reaching grade 鈪,
