舌靜脈注射沒食子酸的慢性低灌注大鼠學(xué)習(xí)記憶能力及腦氧化應(yīng)激損傷相關(guān)指標(biāo)觀察

【摘要】:目的觀察舌靜脈注射沒食子酸(GA)的慢性低灌注大鼠學(xué)習(xí)記憶能力及腦氧化應(yīng)激損傷相關(guān)指標(biāo)變化。方法 48只大鼠隨機分為4組各12只。A、B組大鼠制備慢性低灌注模型,A組在制模前后各5天舌靜脈注射100 mg/kg GA,B組給予等量生理鹽水;C、D組參照A組手術(shù)方法處理大鼠但不結(jié)扎雙側(cè)頸總動脈,C組在手術(shù)前后各5天舌靜脈注射100 mg/kg GA,D組給予等量生理鹽水。比較各組大鼠的學(xué)習(xí)記憶能力(逃避潛伏期、目標(biāo)象限所用時間百分比、游泳速度)及腦氧化應(yīng)激損傷相關(guān)指標(biāo)[超氧化物歧化酶(SOD)、谷胱甘肽過氧化物酶(GSH-Px)、丙二醛(MDA)、總硫醇(TT)]。結(jié)果與D組比較,B組第1~4天逃避潛伏期增加(P均0.05);與B組比較,A組第1~4天逃避潛伏期減少(P均0.05)。與D組比較,B組目標(biāo)象限所用時間百分比減少(P均0.05);與B組比較,A、C組目標(biāo)象限所用時間百分比增加(P均0.05)。與D組比較,B組SOD、GSH-Px、TT降低,MDA升高(P均0.05);與B組比較,A組和C組SOD、GSH-Px、TT升高,MDA降低(P均0.05)。結(jié)論 GA能改善慢性低灌注大鼠的學(xué)習(xí)記憶能力及修復(fù)腦氧化應(yīng)激損傷。
[Abstract]:Objective to observe the changes of learning and memory ability and brain oxidative stress injury in chronic hypoperfusion rats with oral Gallic acid (GA) injection. Methods Forty-eight rats were randomly divided into 4 groups (n = 12 each) and 12 rats in group A (n = 12). But the group C without ligation of bilateral common carotid artery was given the same amount of normal saline 5 days before and after operation. The learning and memory abilities (escape latency, percentage of time used in target quadrant, swimming speed) and the related indexes of brain oxidative stress injury [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malondialdehyde (MDA), total mercaptan (TT)] were compared in each group. Results compared with group D, the escape latency of group B was significantly higher than that of group D (P 0.05), and that of group A was significantly lower than that of group B (P 0.05). Compared with group D, the percentage of time used in target quadrant in group B was decreased (P 0.05), and the percentage of time used in target quadrant in group C was higher than that in group B (P 0.05). Compared with group D, SOD,GSH-Px,TT in group B was lower than that in group D (P 0.05), and SOD,GSH-Px,TT in group A and group C was higher than that in group B (P 0.05). Conclusion GA can improve the ability of learning and memory and repair the oxidative stress injury in chronic hypoperfusion rats.
