豐富環(huán)境對(duì)HIBD新生大鼠海馬CA1區(qū)BDNF及細(xì)胞凋亡的影響
[Abstract]:Aim: to observe the effect of rich environment on BDNF and apoptosis in CA1 area of hippocampus of neonatal HIBD rats and to explore the mechanism of rich environment therapy on HIBD. Methods: the 7-day-old Sprague-Dawley clean newborn rats were randomly divided into three groups: sham operation group, model group and rich environment group. Each group was divided into two subgroups: 14 days after operation and 28 days after operation (nong6). The HIBD animal model of newborn rats was established by reference to the classical Rice-Vannucci method. Model group and sham operation group were fed in standard environment, rich environment group was given rich environment intervention therapy. Morris water maze test was used to detect the learning and memory ability of rats and the positive expression of BDNF protein in hippocampus CA1 area was detected by immunohistochemical method. Tunel method was used to detect apoptosis in CA1 area of hippocampus. The result is 1: 1. Learning and memory ability of rats in each group: at the time point of 14 days and 28 days after operation, compared with the sham operation group, the escape latency of model group was prolonged (P0.05), the number of crossing platform was decreased (P0.05), the difference was statistically significant; compared with model group, The escape latency of rich environment group was shortened (P0.05), the number of crossing platform increased (P0.05), the difference was statistically significant. 2. The expression of BDNF protein in the model group was significantly higher than that in the sham-operated group (P 0.05), and the expression of BDNF protein was significantly higher in the model group than in the sham operation group (P 0.05), and the expression of BDNF protein in the hippocampus of the model group was significantly higher than that in the sham operation group (P 0.05). The positive expression of BDNF protein in hippocampal CA 1 region was more obvious in the rich environment group than in the control group, and the difference was statistically significant (P0.05). 3. The positive expression of apoptotic cells in the model group was significantly higher than that in the sham-operated group at the 28th day after operation (P0.05); compared with the model group, the positive expression of apoptotic cells in the model group was significantly higher than that in the control group (P0.05). The positive expression of apoptotic cells decreased significantly in the rich environment group (P0.05). Conclusion 1. Rich environment can improve learning and memory ability of HIBD rats. 2. 2. Rich environment could up-regulate the expression of BDNF protein in CA 1 area of hippocampus of HIBD rats and down-regulate the expression of apoptotic cells in CA1 area of hippocampus of HIBD rats.
