腦膠質(zhì)瘤中異檸檬酸脫氫酶突變的研究進(jìn)展

【摘要】:異檸檬酸脫氫酶(isocitrate dehvdrogenase,IDH)是三羧酸循環(huán)的關(guān)鍵酶,催化異檸檬酸氧化脫羧生成α-酮戌二酸(α-ketoglutaric acid,α-KG),同時(shí)利用煙酰胺腺嘌呤二核苷酸(nicotinamide adeninedinudeotide,NAD~+)/煙酰胺腺嘌呤二核苷磷酸(nicotinamide adenine dinucleotide phosphate,NADP~+)作為輔助因子生成還原型NADH/NADPH。超過75%的低級別膠質(zhì)瘤(WHO分級為Ⅱ和Ⅲ級)和90%的繼發(fā)性成膠質(zhì)細(xì)胞瘤中存在IDH基因突變,且主要突變位點(diǎn)為R132H(132位精氨酸突變?yōu)榻M氨酸)。IDH基因突變改變了膠質(zhì)瘤的表觀遺傳學(xué)特征,使膠質(zhì)瘤代謝重編程,破壞膠質(zhì)瘤氧化還原穩(wěn)態(tài)。以突變的IDH基因?yàn)橹委煱悬c(diǎn)能抑制膠質(zhì)瘤的進(jìn)展,可用于新的抗腫瘤藥物的研發(fā)。
