天麻素對癲癇大鼠海馬神經(jīng)元IL-1β、TNF-α及IL-10的影響

【摘要】:目的探討IL-1β、TNF-α及IL-10在癲癇發(fā)病中的作用以及天麻素對癲癇大鼠海馬神經(jīng)元IL-1β、TNF-α及IL-10水平的影響。方法將45只成年雄性Wistar大鼠隨機分為正常對照組(NC組)15只、癲癇組(EP組)15只和天麻素組(GE組)15只。EP組和GE組每日1次經(jīng)腹腔注射閾下劑量的戊四氮建立大鼠癲癇模型,然后給予GE組大鼠腹腔注射天麻素,觀察兩組大鼠的行為學(xué)變化。應(yīng)用免疫組織化學(xué)的方法檢測大鼠海馬IL-1β、TNF-α及IL-10水平的變化并進行比較。結(jié)果 GE組的發(fā)作潛伏期較EP組明顯延長,且發(fā)作持續(xù)時間縮短、發(fā)作強度降低(P0.05)。EP組與NC組比較,大鼠海馬神經(jīng)元IL-1β、TNF-α水平明顯上調(diào)(P0.01),而IL-10水平明顯降低(P0.01)。GE組與EP組比較,大鼠海馬神經(jīng)元IL-1β、TNF-α水平明顯降低(P0.01),而IL-10水平明顯升高(P0.01)。結(jié)論 IL-1β、TNF-α可促進癲癇的發(fā)生與發(fā)展,而IL-10有抗癲癇作用;天麻素可通過降低IL-1β、TNF-α水平并升高IL-10而發(fā)揮抗癲癇作用。
[Abstract]:Objective to investigate the role of IL-1 尾 -TNF- 偽 and IL-10 in the pathogenesis of epilepsy and the effect of Gastrodin on the levels of IL-1 尾 -TNF- 偽 and IL-10 in hippocampal neurons of epileptic rats. Methods Forty-five adult male Wistar rats were randomly divided into normal control group (NC group, n = 15), epilepsy group (EP group, n = 15), Gastrodin group (GE group, n = 15) and GE group (n = 15). Then the rats in GE group were given Gastrodin intraperitoneally to observe the behavioral changes of the two groups. The changes of IL-1 尾 -TNF- 偽 and IL-10 in hippocampus of rats were detected by immunohistochemical method and compared. Results compared with NC group, the latency of attack in GE group was significantly prolonged, the duration of attack was shortened and the intensity of attack was decreased (P0.05). Compared with NC group, the level of IL-1 尾 -TNF- 偽 in hippocampal neurons in group GE was significantly up-regulated (P0.01), while the level of IL-10 in group GE was significantly lower than that in group EP (P0.01). The level of IL-1 尾 -TNF- 偽 in hippocampal neurons decreased significantly (P0.01), while the level of IL-10 increased significantly (P0.01). Conclusion IL-1 尾 -TNF- 偽 can promote the occurrence and development of epilepsy, while IL-10 has antiepileptic effect, Gastrodin can play an antiepileptic effect by decreasing the level of IL-1 尾 -TNF- 偽 and increasing IL-10.
