【摘要】：目的探討老年難治性顳葉癲癇患者顳葉組織中神經(jīng)細胞凋亡與信號傳導通路中半胱氨酸天冬氨酸蛋白酶(caspase)-3、caspase-4表達的關(guān)系。方法老年難治性顳葉癲癇患者21例為難治性顳葉癲癇組,術(shù)中均切除顳葉組織;選取同期因腦外傷入院行手術(shù)入路中必須切除的顳葉組織15例為對照組。免疫組化SP法檢測顳葉組織中神經(jīng)細胞caspase-3、caspase-4陽性表達情況;Western印跡檢測顳葉組織中caspase-3、caspase-4蛋白表達情況;TUNEL法檢測兩組神經(jīng)細胞凋亡情況。結(jié)果免疫組化檢測顯示,難治性顳葉癲癇組顳葉組織中caspase-3、caspase-4陽性細胞表達水平明顯高于對照組(P0.05)。Western印跡檢測顯示,難治性顳葉癲癇組顳葉組織中caspase-3、caspase-4蛋白表達量明顯高于對照組(P0.05)。對照組顳葉組織中僅存在少量TUNEL陽性神經(jīng)細胞,難治性顳葉癲癇組中TUNEL陽性神經(jīng)細胞數(shù)明顯增多。難治性顳葉癲癇組細胞凋亡指數(shù)明顯高于對照組(P0.05)。結(jié)論老年難治性顳葉癲癇患者顳葉組織中存在caspase-3、caspase-4傳導的神經(jīng)細胞凋亡,同時caspase-3、caspase-4表達水平與神經(jīng)細胞凋亡呈正相關(guān)。
[Abstract]:Objective to investigate the relationship between neuronal apoptosis and the expression of cysteine aspartate protease (caspase)-3 caspase-4 in the temporal lobe tissues of elderly patients with refractory temporal lobe epilepsy. Methods Twenty-one elderly patients with refractory temporal lobe epilepsy were treated with intractable temporal lobe epilepsy. The expression of caspase-3 and caspase-4 in temporal lobe tissue was detected by immunohistochemical SP method and the expression of caspase-3 and caspase-4 protein in temporal lobe tissue was detected by Western blot. Tunel method was used to detect neuronal apoptosis in both groups. Results Immunohistochemical analysis showed that the expression of caspase-3 and caspase-4 in the temporal lobe of refractory temporal lobe epilepsy group was significantly higher than that in the control group (P0.05). Western blot analysis showed that the expression of caspase-3 and caspase-4 protein in the temporal lobe of refractory temporal lobe epilepsy group was significantly higher than that in the control group (P0.05). There were only a few TUNEL positive neurons in the temporal lobe of the control group, while the number of TUNEL positive neurons in the refractory temporal lobe epilepsy group increased significantly. Apoptosis index in refractory temporal lobe epilepsy group was significantly higher than that in control group (P0.05). Conclusion there is apoptosis of caspase-3 and caspase-4 in the temporal lobe of elderly patients with refractory temporal lobe epilepsy, and the expression level of caspase-3 and caspase-4 is positively correlated with neuronal apoptosis.
