鹽酸法舒地爾對蛛網(wǎng)膜下腔出血大鼠學(xué)習(xí)記憶功能及海馬CA1區(qū)自噬的影響

【摘要】:目的研究鹽酸法舒地爾對蛛網(wǎng)膜下腔出血(SAH)大鼠學(xué)習(xí)記憶及海馬CA1區(qū)自噬的影響。方法雄性Sprague-Dawley大鼠54只,隨機(jī)分為假手術(shù)組、SAH組、藥物組,各18只。頸內(nèi)動脈刺破法造模,藥物組造模成功后予鹽酸法舒地爾10 mg/kg腹腔注射,每天1次,假手術(shù)組和SAH組腹腔注射等量生理鹽水,干預(yù)后6 h、24 h、72 h予穿梭箱試驗(yàn);取海馬CA1區(qū)組織HE染色觀察細(xì)胞形態(tài),免疫組織化學(xué)染色觀察Beclin-1和微管相關(guān)蛋白l輕鏈3-Ⅱ(LC3-Ⅱ)的表達(dá)。結(jié)果與假手術(shù)組相比,SAH組大鼠各時(shí)間點(diǎn)穿梭箱試驗(yàn)逃避反應(yīng)次數(shù)減少(P0.05),逃避反應(yīng)時(shí)間延長(P0.05);海馬CA1區(qū)神經(jīng)細(xì)胞數(shù)目減少(P0.05),Beclin-1、LC3-Ⅱ表達(dá)增高(P0.05)。與SAH組相比,藥物組各時(shí)間點(diǎn)逃避反應(yīng)次數(shù)增多(P0.05),逃避反應(yīng)時(shí)間減少(P0.05);海馬CA1區(qū)神經(jīng)細(xì)胞增多(P0.05),Beclin-1、LC3-Ⅱ表達(dá)進(jìn)一步增多(P0.05)。結(jié)論鹽酸法舒地爾可改善SAH大鼠學(xué)習(xí)記憶功能,減少神經(jīng)細(xì)胞損傷,增強(qiáng)海馬CA1區(qū)自噬。
