新生小鼠缺氧缺血性腦損傷模型的建立

【摘要】:目的對經(jīng)典Vannucci法進行改進,建立一種簡便、穩(wěn)定的新生小鼠缺血缺氧腦損傷模型。方法將新生11 d的KM小鼠分為正常對照組(N組,n=20)和缺血缺氧組(HIBD組,n=160),對HIBD組行左側(cè)頸總動脈結(jié)扎,分別按照C1-C8條件缺氧建模。建模后通過比較各條件下小鼠死亡率、建模成功率和TTC染色腦梗死體積,選取最穩(wěn)定的建模條件。建模后利用體重生長曲線分析小鼠生長發(fā)育情況;Longa、Grip test、懸吊試驗評估小鼠神經(jīng)運動功能;HE染色觀察腦組織病理改變。結(jié)果新生小鼠行左側(cè)頸總動脈結(jié)扎,8% O_2、35℃條件下缺氧45min,死亡率低(8.3%)且成模率高(47.92%);HIBD組較N組小鼠體重增長緩慢并出現(xiàn)嚴重的神經(jīng)運動功能障礙;結(jié)扎側(cè)出現(xiàn)腦梗死區(qū),約占全腦體積(17.76±0.70)%;結(jié)扎側(cè)大腦皮層及海馬區(qū)神經(jīng)元出現(xiàn)變性壞死。結(jié)論本實驗采用新生小鼠行左側(cè)頸總動脈結(jié)扎,在8%O2、35℃條件下缺氧45 min復(fù)制HIBD動物模型,簡便且穩(wěn)定性好,是用于新生兒缺血缺氧性腦損傷研究較為理想的動物模型。
[Abstract]:Objective to improve the classical Vannucci method and to establish a simple and stable model of cerebral ischemia and hypoxia injury in newborn mice. Methods the km mice were divided into two groups: normal control group (N group) and ischemic hypoxia group (HIBD group). The left common carotid artery was ligated in HIBD group, and the hypoxia model was established according to C1-C8 condition. After modeling, the most stable modeling conditions were selected by comparing the mortality of mice, the success rate of modeling and the volume of cerebral infarction stained by TTC. After modeling, the growth and development of mice were analyzed by weight growth curve, and the neuromotor function of mice was evaluated by suspension test. The pathological changes of brain tissue were observed by HE staining. Results the neonatal mice underwent left common carotid artery ligation (8%) for 45 min under hypoxia, the mortality rate was low (8.3%) and the rate of model formation was higher (47.92%) in HIBD group than that in N group, the body weight of HIBD group increased more slowly than that of N group, and severe neuromotor dysfunction occurred in ligated side, and cerebral infarction area appeared in ligation side. The volume of the whole brain was (17.76 鹵0.70) and the neurons in the cortical and hippocampal regions were denatured and necrotized.
