【摘要】：目的：研究分析五味子的有效成分五味子木脂素對(duì)大鼠癲癇模型的抗癲癇作用及神經(jīng)保護(hù)。 方法：Wistar大鼠90只，分為三組，對(duì)照組10只（不予特殊處理），癲癇模型組40只，五味子木脂素組40只，腹腔注射東莨菪堿硝酸甲酯(1mg/kg),20分鐘后給予匹羅卡品（320-350mg/kg,腹腔注射）首先誘導(dǎo)大鼠癲癇持續(xù)狀態(tài)發(fā)作，誘導(dǎo)成功后予以地西泮5mg/kg肌肉注射，終止發(fā)作。隨后1個(gè)月（30天）用視頻記載大鼠癲癇發(fā)作次數(shù)及發(fā)作過(guò)程，1個(gè)月內(nèi)發(fā)作3次以上為造模成功。五味子木脂素組給予五味子木脂素80mg/kg喂養(yǎng)，對(duì)照組及癲癇組正常大鼠飲食。視頻記錄三組大鼠的癲癇發(fā)作情況。記錄完成后，處死并灌注取腦，腦組織按重量體積比1：9加入4℃生理鹽水，，制備10%的勻漿。應(yīng)用分光光度法，考馬斯亮蘭法定蛋白。檢測(cè)記錄三組大鼠血清、大腦組織中的NO、T-AOC、SOD、MDA含量。分析五味子木脂素對(duì)癲癇模型大鼠的神經(jīng)保護(hù)作用。 結(jié)果:應(yīng)用視頻記錄1個(gè)月內(nèi)上述三組大鼠癲癇發(fā)作情況。視頻記錄結(jié)果顯示：五味子木脂素組40只鼠有17只出現(xiàn)癲癇發(fā)作（發(fā)作率為42.5%），癲癇組33只出現(xiàn)癲癇發(fā)作（82.5%），對(duì)照組0只出現(xiàn)癲癇發(fā)作（0%），結(jié)果顯示五味子木脂素具有明顯減少癲癇發(fā)作次數(shù)的作用。五味子木脂素組大鼠血液及腦組織中SOD和T-AOC活性增高， MDA、NO含量明顯降低。 結(jié)論: 1.五味子木脂素明顯減少癲癇發(fā)作次數(shù)。 2.五味子木脂素通過(guò)減少自由基的產(chǎn)生及抗氧化作用對(duì)癲癇腦組織具有保護(hù)作用。
[Abstract]:Aim: to study the antiepileptic effect and neuroprotective effect of Schisandra chinensis lignin on rat epileptic model. Methods 90 Wistar rats were divided into three groups: control group (n = 10), epileptic model group (n = 40) and Schisandrin group (n = 40). After 20 minutes of intraperitoneal injection of scopolamine methyl nitrate (1mg/kg), pilocarpine (320-350 mg / kg, intraperitoneal injection) was used to induce epileptic persistent state seizure in rats. After successful induction, diazepam 5mg/kg was injected intramuscularly to terminate the seizure. After 1 month (30 days), the number and process of epileptic seizures in rats were recorded by video, and more than 3 seizures in one month were successfully established. Schisandrin group was fed with Schisandrin 80mg/kg, control group and epileptic group were fed with normal diet. The epileptic seizures were recorded in three groups of rats.
