特制牛肉湯對(duì)腦缺血大鼠外周血白細(xì)胞介素-1β和干擾素-γ的影響

【摘要】:目的研究特制牛肉湯對(duì)腦缺血大鼠外周血白細(xì)胞介素(IL)-1β及干擾素(IFN)-γ的影響。方法將96只雄性SD大鼠按隨機(jī)數(shù)字表分為假手術(shù)組、模型組、特制牛肉湯組和肌肽組,每組24只。模型組、特制牛肉湯組和肌肽組采用線栓法制備永久性大腦中動(dòng)脈閉塞模型(p MCAO),于術(shù)后清醒2 h內(nèi)首次灌胃給藥,特制牛肉湯組予600 mg·kg~(-1)·d~(-1)特制牛肉湯,肌肽組予600 mg·kg~(-1)·d~(-1)肌肽水溶液。假手術(shù)組與模型組予等量無菌生理鹽水,每日2次,各組大鼠分別于造模后1 d,3 d和7 d處死,各組各時(shí)點(diǎn)8只大鼠。采用Longa評(píng)分法進(jìn)行神經(jīng)功能缺損評(píng)分,2,3,5-氯化三苯基四氮唑(TTC)染色法測(cè)定腦梗死體積分?jǐn)?shù)、酶聯(lián)免疫吸附法(ELISA)測(cè)外周血中IL-1β、IFN-γ表達(dá)。結(jié)果腦缺血后,各時(shí)點(diǎn)模型組大鼠神經(jīng)功能評(píng)分顯著上升;與模型組相比,予特制牛肉湯7 d后,腦缺血大鼠神經(jīng)功能評(píng)分顯著下降(P0.05)。腦缺血后,模型組、特制牛肉湯組和肌肽組均可見大腦中動(dòng)脈供血區(qū)梗死灶。與模型組相比,予特制牛肉湯和肌肽1 d和3 d后,腦梗死體積分?jǐn)?shù)有下降趨勢(shì),但無統(tǒng)計(jì)學(xué)差異(P0.05);予特制牛肉湯7 d后,腦梗死體積分?jǐn)?shù)顯著降低(P0.01)。腦缺血后,各時(shí)點(diǎn)模型組大鼠外周血IL-1β水平顯著上升(P0.05),與模型組相比,予特制牛肉湯7 d后,IL-1β水平明顯下降(P0.05);予肌肽7 d后,IL-1β有下降趨勢(shì),但無統(tǒng)計(jì)學(xué)差異(P0.05)。腦缺血后3 d和7 d,模型組大鼠外周血IFN-γ水平顯著上升(P0.05),與模型組相比,予特制牛肉湯和肌肽7 d后,IFN-γ水平明顯下降(P0.01)。結(jié)論特制牛肉湯通過下調(diào)促炎細(xì)胞因子IL-1β、IFN-γ表達(dá),從而減輕腦缺血大鼠炎癥反應(yīng),改善神經(jīng)功能缺損癥狀,對(duì)腦起保護(hù)作用。
[Abstract]:Objective to study the effects of special beef decoction on interleukin (IL)-1 尾 and interferon (IFN)-緯 in peripheral blood of rats with cerebral ischemia. Methods 96 male Sprague-Dawley rats were randomly divided into sham-operation group, model group, special beef soup group and carnosine group with 24 rats in each group. Model group, special beef decoction group and carnosine group were used to establish permanent middle cerebral artery occlusion model (pMCAO) by thread occlusion. The rats in the special beef decoction group were given 600 mg kg ~ (-1) d ~ (-1) special beef soup, and the carnosine group were given 600 mg kg ~ (-1) d ~ (-1) carnosine solution. The sham-operation group and the model group were given the same amount of aseptic saline twice a day. The rats in each group were killed on the 1st day and 7th day, respectively. There were 8 rats in each group at each time point. The neurological function defect score (NDS) was measured by Longa score method. The volume fraction of cerebral infarction was measured by TTC staining and the expression of IL-1 尾 -IFN- 緯 in peripheral blood was measured by enzyme linked immunosorbent assay (Elisa). Results after cerebral ischemia, the neurological function score of the model group increased significantly, compared with the model group, after 7 days of special beef decoction, the neurological function score of the cerebral ischemic rats decreased significantly (P0.05). After cerebral ischemia, the infarcts of the middle cerebral artery were found in the model group, the special beef decoction group and the carnosine group. Compared with the model group, the volume fraction of cerebral infarction decreased after 1 and 3 days of treatment with special beef soup and carnosine (P0.05), and the volume fraction of cerebral infarction decreased significantly after 7 days (P0.01). After cerebral ischemia, the level of IL-1 尾 in peripheral blood of the model group increased significantly (P0.05), compared with the model group, the level of IL-1 尾 decreased significantly after 7 days of treatment with special beef decoction (P0.05), and the level of IL-1 尾 decreased after 7 days of treatment with carnosine, but there was no statistical difference (P0.05). The level of IFN- 緯 in peripheral blood of the model group increased significantly 3 days and 7 days after cerebral ischemia (P0.05). Compared with the model group, the level of IFN- 緯 in the model group decreased significantly after 7 days of treatment with special beef decoction and carnosine (P0.01). Conclusion the special beef decoction can reduce the inflammatory reaction, improve the symptom of nerve function defect and protect the brain by down-regulating the expression of IL-1 尾 -IFN- 緯 in rats with cerebral ischemia.
