惰性氣體神經(jīng)保護(hù)作用的行為學(xué)研究

【摘要】:目的利用乳鼠缺血缺氧模型,探討腦損傷動(dòng)物模型的基本生長(zhǎng)發(fā)育情況、協(xié)調(diào)運(yùn)動(dòng)能力和記憶功能,同時(shí)對(duì)幼年時(shí)期給予惰性氣體暴露能否改善動(dòng)物模型的長(zhǎng)期神經(jīng)行為功能進(jìn)行考察。方法結(jié)扎出生后7d的Sprague-Dawley大鼠的右頸總動(dòng)脈,1h后給予體積分?jǐn)?shù)為0.08的氧氣缺氧處理90min,2h后分別給予體積分?jǐn)?shù)為0.7的氬氣、氦氣和氙氣(氬氣組、氦氣組和氙氣組,每組20只)或空氣(陽(yáng)性對(duì)照組,20只)處理90min,其間將氣體暴露罐置于水浴中保持37℃恒溫;另有1組乳鼠未經(jīng)任何處理(陰性對(duì)照組,20只)。將各組大鼠喂養(yǎng)至出生后38d時(shí)分別進(jìn)行平衡疲勞轉(zhuǎn)動(dòng)棒實(shí)驗(yàn)、恐懼條件反射實(shí)驗(yàn)和Morris水迷宮實(shí)驗(yàn)。結(jié)果氦氣組大鼠出生后38d時(shí)的體重顯著輕于陰性對(duì)照組和陽(yáng)性對(duì)照組(P值均0.01),氬氣組和氙氣組大鼠出生后38d時(shí)的體重與陰性對(duì)照組和陽(yáng)性對(duì)照組間的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05)。平衡疲勞轉(zhuǎn)動(dòng)棒實(shí)驗(yàn)中,轉(zhuǎn)速為30r/min時(shí),5組大鼠潛伏期的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05);轉(zhuǎn)速增加至50r/min時(shí),陽(yáng)性對(duì)照組的潛伏期顯著短于其他4組同轉(zhuǎn)速時(shí)和同組轉(zhuǎn)速為30r/min時(shí)(P值均0.01),其他4組間潛伏期的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05)?謶謼l件反射實(shí)驗(yàn)中,實(shí)驗(yàn)第2天的環(huán)境測(cè)試中,陽(yáng)性對(duì)照組大鼠的靜止呆滯時(shí)間百分比顯著小于其他4組(P值分別0.01、0.05);實(shí)驗(yàn)第3天的聲音刺激測(cè)試中,無(wú)論是在適應(yīng)期還是之后的每次聲音刺激后,5組大鼠間靜止呆滯時(shí)間百分比的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05)。水迷宮實(shí)驗(yàn)第1天,5組大鼠間尋找平臺(tái)潛伏期的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05),第2天起,除陽(yáng)性對(duì)照組外,其他4組大鼠的潛伏期明顯縮短,第3~5天,陽(yáng)性對(duì)照組大鼠的潛伏期顯著長(zhǎng)于其他4組大鼠同時(shí)間(P值均0.05),其他4組大鼠間的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05);由AUC表示的尋找平臺(tái)潛伏期的累積時(shí)間提示,陽(yáng)性對(duì)照組大鼠尋找平臺(tái)潛伏期顯著長(zhǎng)于陰性對(duì)照組和氬氣組、氦氣組、氙氣組(P值分別0.01、0.05);5組大鼠間平均游泳速度的差異均無(wú)統(tǒng)計(jì)學(xué)意義(P值均0.05)。結(jié)論氬氣、氦氣和氙氣對(duì)乳鼠缺血缺氧腦損傷模型均具有神經(jīng)保護(hù)功能,氦氣因其可能影響乳鼠全身發(fā)育生長(zhǎng)而需進(jìn)一步研究。氬氣作為一種便宜且較易獲得的惰性氣體,具有良好的神經(jīng)保護(hù)劑的應(yīng)用前景。
[Abstract]:Objective to investigate the basic growth and development of brain injury animal model and to coordinate motor ability and memory function by using the model of ischemia and hypoxia in neonatal rats.
