【摘要】：目的檢測人頸動脈粥樣硬化斑塊組織及正常頸動脈組織中去乙酰化酶3(SIRT3)的表達(dá)水平,分析其與頸動脈粥樣硬化斑塊形成的關(guān)系及臨床意義。方法利用HE染色、Masson’s trichrome染色及油紅O染色對6例頸動脈組織進(jìn)行粥樣硬化斑塊分期,利用免疫組化法和Western blot檢測不同分期的斑塊組織及正常頸動脈組織中SIRT3的蛋白表達(dá)差異,利用免疫熒光共定位法觀察SIRT3的細(xì)胞定位。結(jié)果 6例組織分別為2例正常血管組織,2例Ⅰ期動脈粥樣硬化組織,2例Ⅳ期動脈粥樣硬化斑塊組織,Western blot及免疫組化染色顯示SIRT3在Ⅳ期頸動脈粥樣硬化斑塊組織中顯著表達(dá),正常血管組織及Ⅰ期粥樣硬化血管組織中未見SIRT3表達(dá);免疫熒光共定位染色結(jié)果顯示,SIRT3在Ⅳ期斑塊內(nèi)的內(nèi)皮細(xì)胞、平滑肌細(xì)胞和巨噬細(xì)胞內(nèi)均有顯著表達(dá)。結(jié)論SIRT3在頸動脈粥樣硬化Ⅳ期斑塊中表達(dá)顯著增高,可能作為評估粥樣硬化穩(wěn)定性的一個重要指標(biāo)。
[Abstract]:Objective to detect the expression of deacetylase 3 (SIRT3) in human carotid atherosclerotic plaque and normal carotid artery, and to analyze its relationship with carotid atherosclerotic plaque formation and its clinical significance. Methods six cases of carotid atherosclerotic plaques were staging with HE staining and oil red O staining. The protein expression of SIRT3 was detected by immunohistochemistry and Western blot.
