芹黃素抑制小膠質(zhì)細(xì)胞活化作用研究

【摘要】:目的探討芹黃素對(duì)小膠質(zhì)細(xì)胞活化的抑制作用。方法原代培養(yǎng)SD大鼠小膠質(zhì)細(xì)胞,實(shí)驗(yàn)隨機(jī)分為空白對(duì)照組、脂多糖組(LPS組)、LPS+芹黃素(10μM)組、LPS+芹黃素(20μM)組、LPS+芹黃素(50μM)組。MTT法檢測(cè)芹黃素對(duì)小膠質(zhì)細(xì)胞活性的影響;ELISA法檢測(cè)IL-1、IL-10等炎癥相關(guān)因子以及BDNF、PDNF等神經(jīng)營(yíng)養(yǎng)因子的表達(dá);RTPCR、western blot檢測(cè)炎癥相關(guān)基因i NOS轉(zhuǎn)錄以及表達(dá)水平。結(jié)果 MTT檢測(cè)結(jié)果表明,芹黃素對(duì)小膠質(zhì)細(xì)胞活性無(wú)明顯抑制。LPS刺激后,芹黃素預(yù)處理組IL-1等炎癥因子表達(dá)水平明顯低于單獨(dú)LPS組(P0.05);而芹黃素預(yù)處理組IL-10的表達(dá)則高于單獨(dú)LPS組(P0.01);與此同時(shí),BDNF、PDNF等神經(jīng)營(yíng)養(yǎng)因子的分泌也有相同趨勢(shì)(P0.05)。芹黃素預(yù)處理組i NOS表達(dá)和轉(zhuǎn)錄水平也低于單獨(dú)LPS組(P0.05)。結(jié)論芹黃素可抑制活化狀態(tài)小膠質(zhì)細(xì)胞炎癥因子以及炎癥相關(guān)蛋白的表達(dá),并可同時(shí)促進(jìn)與神經(jīng)元分化成熟相關(guān)神經(jīng)營(yíng)養(yǎng)因子的分泌。
[Abstract]:Objective to investigate the inhibitory effect of celanthin on the activation of microglia. Methods Sprague-Dawley rat microglia cells were cultured in primary culture and were randomly divided into blank control group. Effects of lipopolysaccharide (LPS) on lipopolysaccharide (LPS) group (10 渭 M) lipopolysaccharide (LPS) celanthin (20 渭 M) group (lipopolysaccharide) group (50 渭 M). MTT assay was used to detect the effect of celanthin on microglial activity. Elisa was used to detect the expression of inflammatory related factors such as IL-1, IL-10 and neurotrophic factors such as BDNF PDNF by RT PCR western blot. I NOS transcription and expression level were measured. Results the results of MTT showed that celanthin did not inhibit the activity of microglia. The expression of IL-1 and other inflammatory factors were significantly lower in the celanthin pretreatment group than in the LPS group, while the expression of IL-10 in the carvingin preconditioning group was higher than that in the LPS group, and there was a similar trend in the secretion of neurotrophic factors such as P0.05 in the LPS preconditioning group. The expression and transcription level of I NOS in celanthin preconditioning group was also lower than that in LPS group alone (P 0.05). Conclusion celanthin can inhibit the expression of inflammatory cytokines and inflammatory related proteins in activated microglia and promote the secretion of neurotrophic factors related to neuronal differentiation and maturation at the same time.
