【摘要】：目的探討腦膜瘤組織中PRDX4的表達(dá)差異及其與臨床病理的相關(guān)性。方法采用Western印跡和免疫組化(SP法)分析68例腦膜瘤組織、59例配對(duì)的瘤旁腦膜組織及45例正常腦膜組織中PRDX4的表達(dá)情況,并分析腦膜瘤組織中PRDX4與核因子(NF)-κB活化產(chǎn)物p65的相關(guān)性,進(jìn)一步應(yīng)用分層分析、生存分析和Cox多元回歸分析腦膜瘤患者PRDX4的表達(dá)與臨床病理及其預(yù)后的關(guān)系。結(jié)果腦膜瘤組織PRDX4蛋白條帶灰度值較瘤旁和正常腦膜組織顯著增高(P0.001),且瘤旁腦膜組織PRDX4蛋白表達(dá)較正常腦膜組織高(P0.05);腦膜瘤PRDX4與NF-κB p65蛋白表達(dá)具有顯著正相關(guān)性(P0.05);且腦膜瘤患者中PRDX4陽(yáng)性表達(dá)與臨床分期有顯著相關(guān)性(P0.01);進(jìn)一步生存分析發(fā)現(xiàn)PRDX4表達(dá)陽(yáng)性者5年無瘤生存期和生存率顯著低于陰性者(P0.05),且Cox分析示PRDX4可能是腦膜瘤患者5年無瘤生存預(yù)后的獨(dú)立影響因素(P0.05;RR=1.484;95%CI:1.077~2.260)。結(jié)論 NF-κB激活調(diào)控蛋白PRDX4可能為早期診斷腦膜瘤的生物學(xué)標(biāo)志,且與腦腫瘤臨床分期密切相關(guān),可能具有獨(dú)立預(yù)測(cè)腦膜瘤患者5年無瘤生存預(yù)后的重要臨床價(jià)值。
[Abstract]:Objective to investigate the expression of PRDX4 in meningiomas and its correlation with clinicopathology. Methods Western blot and immunohistochemical staining were used to analyze the expression of PRDX4 in 59 matched meningioma tissues and 45 normal meningeal tissues, and the correlation between PRDX4 and NF- 魏 B activator p65 in meningiomas. Furthermore, stratified analysis, survival analysis and Cox multivariate regression analysis were used to analyze the relationship between the expression of PRDX4 and clinicopathology and prognosis in meningioma patients. Results the gray level of PRDX4 protein bands in meningiomas was significantly higher than that in adjacent and normal meninges, and the expression of PRDX4 protein in meningiomas was significantly higher than that in normal meningeal tissues, and there was a significant positive correlation between PRDX4 and NF- 魏 B p65 protein expression in meningiomas. The positive expression of PRDX4 was significantly correlated with the clinical stage in meningioma patients. Further survival analysis showed that the 5-year tumor-free survival time and survival rate of patients with positive PRDX4 expression were significantly lower than that of patients with negative PRDX4 expression, and Cox analysis showed that PRDX4 might be meningeal. The independent factors influencing the survival and prognosis of tumor-free patients in 5 years were P0.05 RRX 1.48495% CI: 1.07772.260. Conclusion the NF- 魏 B activator protein PRDX4 may be a biological marker for early diagnosis of meningioma and closely related to the clinical stage of meningioma. It may have an important clinical value in predicting the survival and prognosis of meningioma patients in 5 years without tumor.
