脊髓自噬功能與糖尿病神經(jīng)病理性疼痛的相關(guān)性研究

【摘要】:目的探討脊髓自噬功能與糖尿病神經(jīng)病理性疼痛(DNF)的相關(guān)性。方法選取STZ制備的糖尿病模型,隨機(jī)分為制模后3 d組、制模后1周組、制模后2周組、制模后3周組及制模后4周組,每組20只。同期選取5組健康大鼠分為對照1~5組(Con 1~5),每組20只,注射檸檬酸緩沖液。比較各組血糖、機(jī)械疼痛閾值、熱刺疼痛閾值,以及脊髓自噬功能和脊髓補(bǔ)體C3的表達(dá)。結(jié)果制模后各組血糖均高于同期對應(yīng)的Con組(P0.05),除制模后3 d組外,其余各組機(jī)械疼痛閾值低于同期對應(yīng)的Con組(P0.05)。制模后3周組、制模后4周組熱刺激疼痛閾值也低于同期對應(yīng)的Con組(P0.05)。制模后2周組、制模后3周組和制模后4周組C3 mRNA、補(bǔ)體C3、Beclin-Ⅰ和LC3-Ⅱ表達(dá)均高于同期對應(yīng)的Con組(P0.05)。制模后3 d組、制模后1周組、制模后2周組、制模后3周組和制模后4周組P62蛋白表達(dá)均低于同期對應(yīng)的Con組(P0.05)。結(jié)論大鼠脊髓補(bǔ)體C3、Beclin-Ⅰ、LC3-Ⅱ、P62激活提示脊髓自噬功能激活;脊髓自噬功能激活可能參與了DNP發(fā)生發(fā)展。
[Abstract]:Objective to investigate the correlation between spinal cord autophagy and diabetic neuropathic pain (DNF). Methods Diabetic model made by STZ was randomly divided into three groups, one week after model making, two weeks after modeling, three weeks after modeling and four weeks after making model, with 20 rats in each group. At the same time, five groups of healthy rats were divided into control group (n = 20) and control group (n = 20). Blood glucose, mechanical pain threshold, Tottene pain threshold, spinal autophagy and C3 expression in spinal cord were compared. Results the blood glucose in each group was higher than that in Con group at the same time, and the mechanical pain threshold of other groups was lower than that of Con group in the same period except for 3 d after model making. The pain threshold of thermal stimulation in the group of 3 weeks after modeling and 4 weeks after modeling was also lower than that in the corresponding Con group (P 0.05).
