S-腺苷蛋氨酸通過降低Cav-1的表達保護KCl刺激引起的神經(jīng)元損傷

【摘要】:目的探討S-腺苷蛋氨酸(S-adenosylmethionine,SAM)對KCl刺激引起的原代培養(yǎng)的小鼠皮層神經(jīng)元損傷是否有保護作用及機制。方法將原代培養(yǎng)1周的小鼠皮層神經(jīng)元隨機分為3組:正常(Control)組、KCl組、SAM-KCl組。用倒置顯微鏡觀察各組細胞形態(tài)的變化,用q PCR、Western blot分別檢測各組質(zhì)膜微囊結(jié)構(gòu)蛋白~(-1)(caveolin~(-1),Cav~(-1))mRNA及蛋白的表達變化。結(jié)果 0.05 mol·L~(-1)KCl作用于原代培養(yǎng)1周的小鼠皮層神經(jīng)元12 h可使神經(jīng)元產(chǎn)生明顯的損傷,突起縮短變細,胞體變小,細胞聚團。0.02 mol·L~(-1)SAM預(yù)處理6 h可明顯降低KCl引起的細胞損傷,q PCR、Western blot分析顯示KCl可使原代培養(yǎng)1周的皮層神經(jīng)元Cav~(-1) mRNA及蛋白表達增高(P0.01),SAM可降低KCl引起的神經(jīng)元Cav~(-1) mRNA及蛋白表達增高(P0.05)。結(jié)論 SAM對KCl引起的神經(jīng)元損傷有保護作用,該保護作用與其降低Cav~(-1)的表達有關(guān)。
[Abstract]:Objective to investigate the protective effect and mechanism of S-adenosylmethionine (S-adenosylmethionine) on primary cultured cortical neuron injury induced by KCl in mice. Methods the primary cultured cortical neurons were randomly divided into three groups: normal control group (KCl) group and SAM-KCl group. The morphologic changes of the cells in each group were observed by inverted microscope. The mRNA and protein expression of the plasma membrane microencapsulation protein (PMC) were detected by qPCRX Western blot in each group. Results the effects of 0. 05 mol L~(-1)KCl on primary cultured mouse cortical neurons for 12 h resulted in obvious damage to the neurons, the process was shortened and the body of the neurons became smaller. Cell cluster. 0.02 mol L~(-1)SAM pretreatment for 6 h significantly reduced cell damage induced by KCl. Western blot analysis showed that KCl could increase the expression of mRNA and protein in primary cultured cortical neurons for 1 week. Conclusion SAM has a protective effect on the neuronal injury induced by KCl, which is related to the decrease of Cavo 1 expression.
