【摘要】:目的通過(guò)觀察福爾馬林致炎大鼠鞘內(nèi)給藥后的行為學(xué)變化,探討星形膠質(zhì)細(xì)胞內(nèi)的三羧酸循環(huán)在福爾馬林誘導(dǎo)的脊髓中樞敏化中的作用。 方法建立炎性痛動(dòng)物模型,通過(guò)行為學(xué)檢測(cè)觀察了福爾馬林(5%,50μ1)致炎大鼠鞘內(nèi)給予1nmol/10μl氟代檸檬酸(fluorocitrate,FC)和(或)500nomol/10μl谷氨酸(L-glutamate,Glu)后大鼠行為學(xué)變化情況。具體如下:(1)福爾馬林炎性持續(xù)性痛模型:選擇鞘內(nèi)置管術(shù)后5d恢復(fù)良好的大鼠右后肢足底中心皮下注射(s.c.)5%的福爾馬林50μl。(2)鞘內(nèi)給藥:a.福爾馬林炎性持續(xù)性痛急性期:對(duì)照組:福爾馬林注射前20分鐘,分別鞘內(nèi)注射①12%DMSO(n=3)②谷氨酸(500nmol/10μl,n=3);實(shí)驗(yàn)組:③福爾馬林注射前20分鐘,鞘內(nèi)注射FC(1nmol/10μl, n=6),④鞘內(nèi)注射500nmol/10μl Glu10分鐘后,再次鞘內(nèi)注射1nmol/10μl FC,20分鐘后右后肢足底注射福爾馬林(n=6)。推進(jìn)速度為0.5μl/s,注射福爾馬林后即刻將實(shí)驗(yàn)大鼠放入實(shí)驗(yàn)箱進(jìn)行行為學(xué)測(cè)試。b.福爾馬林炎性持續(xù)性痛慢性期:①福爾馬林注射前20分鐘,分別鞘內(nèi)注射12%DMSO(n=3).FC(1nmol/10μl,n=6),并在福爾馬林注射后3h、6h、1d、2d、3d、4d進(jìn)行行為學(xué)檢測(cè);②福爾馬林注射前20分鐘,注射后1d,2d,連續(xù)3次分別鞘內(nèi)注射12%DMSO(n=3).FC(1nmol/10μl, n=6),并在福爾馬林注射后1d、2d、3d、5d、7d、9d、14d進(jìn)行行為學(xué)檢測(cè);③福爾馬林注射后9d,分別鞘內(nèi)注射12%DMS0(n=3).FC(1nmol/10μl, n=6),并在福爾馬林注射后1d、3d、5d、7d、9d(0h)、9d(3h)、9d(6h)、10d、11d、14d進(jìn)行行為學(xué)檢測(cè);④福爾馬林注射后9d、10d、11d,分別鞘內(nèi)注射12%DMSO(n=3).FC(1nmol/10μ l,n=6),并在福爾馬林注射后1d、3d、5d、7d、9d、10d、11d、12d、13d、14d進(jìn)行行為學(xué)檢測(cè)。 結(jié)果(1)鞘內(nèi)給予FC對(duì)炎性痛大鼠自發(fā)傷害性行為(舔咬爪和縮腿反射)有抑制作用,而鞘內(nèi)給予Glu對(duì)此作用產(chǎn)生部分的翻轉(zhuǎn)效應(yīng)。 (2)在急性期:與對(duì)照組相比,單次(福爾馬林注射20min前)鞘內(nèi)給予FC能夠在1-3內(nèi)抑制大鼠的機(jī)械痛敏,并在第1天顯著抑制大鼠機(jī)械痛敏(P≤0.01);多次(福爾馬林注射20min前和注射后1d、2d連續(xù)3天)鞘內(nèi)注射FC能夠在1-7天內(nèi)抑制大鼠的同側(cè)和對(duì)側(cè)的機(jī)械痛敏,在第5天顯著抑制大鼠同側(cè)機(jī)械痛敏(P≤0.01),在第3天顯著抑制大鼠對(duì)側(cè)機(jī)械痛敏(P≤0.01)。在慢性期:與對(duì)照組相比,單次(福爾馬林注射后第9天)鞘內(nèi)注射FC3小時(shí)時(shí)能夠抑制大鼠的同側(cè)和對(duì)側(cè)的機(jī)械痛敏(P≤0.05),但在6小時(shí)時(shí)其機(jī)械痛敏恢復(fù);多次(福爾馬林注射后9d、l0d、l1d連續(xù)3天)鞘內(nèi)注射FC能夠?qū)ν瑐?cè)和對(duì)側(cè)的機(jī)械痛敏產(chǎn)生瞬時(shí)的抑制的作用。 結(jié)論鞘內(nèi)注射氟代檸檬酸能顯著降低炎性痛大鼠的自發(fā)傷害性行為及誘發(fā)傷害性行為,而谷氨酸能夠部分翻轉(zhuǎn)此作用,提示星形膠質(zhì)細(xì)胞內(nèi)三羧酸循環(huán)在脊髓中樞敏化中起著重要作用。
[Abstract]:Aim to investigate the role of tricarboxylic acid cycle in astrocytes in formalin-induced spinal central sensitization by observing the behavioral changes after intrathecal administration of formalin-induced inflammatory rats.
