【摘要】：目的研究缺血性腦血管病(ICVD)核因子(NF)-κB對腫瘤壞死因子(TNF)-α表達(dá)的影響及其對大鼠腦組織損傷的作用。方法將48只Wistar大鼠隨機(jī)分成正常組(12只)、模型組(12只)、假手術(shù)組(12只)和吡咯烷二硫基甲酸酯(PDTC)+模型組,每組12只。采用線栓法制成大鼠局部缺血再灌注模型,用Ludmila Belayev 12分評分法評估大鼠神經(jīng)功能,氯化-2,3,5-三苯基四氮唑(TTC)染色法觀察腦梗死體積,免疫組化法檢測腦組織NF-κB表達(dá),酶聯(lián)免疫吸附法(ELISA)測腦組織TNF-α含量。結(jié)果模型組大鼠Ludmila Belayev 12得分明顯高于正常組與假手術(shù)組(P0.05),PDTC+模型組得分顯著低于模型組(P0.01);模型組大鼠的腦梗死體積顯著多于正常組和假手術(shù)組(P0.01),與模型組比較,PDTC+模型組大鼠腦梗死體積顯著減少(P0.05);模型組大鼠腦組織NF-κB陽性細(xì)胞表達(dá)率顯著高于正常組和假手術(shù)組(P0.05),DPTC+模型組NF-κB表達(dá)明顯低于模型組(P0.05);模型組大鼠TNF-α的含量顯著高于正常組和假手術(shù)組(P0.01),PDTC+模型組TNF-α含量明顯低于模型組(P0.05)。結(jié)論 PDTC抑制NF-κB活性可以減少TNF-α含量,降低TNF-α的表達(dá),所以可通過NF-κB調(diào)節(jié)TNF-α表達(dá)對缺血再灌注腦損傷起到保護(hù)作用,可能與其抑制炎癥反應(yīng)作用有關(guān)。
[Abstract]:Objective to study the effect of NF-魏 B on the expression of TNF- 偽 and its effect on brain tissue injury in rats with ischemic cerebrovascular disease (ICVD). Methods 48 Wistar rats were randomly divided into normal group (n = 12), model group (n = 12), sham operation group (n = 12) and pyrrolidine dithioformate group (n = 12). The rat model of local ischemia reperfusion was established by the method of thread embolization. The nerve function was evaluated by Ludmila Belayev 12 score method, the infarct volume was observed by chlorination and the expression of NF- 魏 B in brain tissue was detected by immunohistochemical method, and the cerebral infarct volume was observed by using the method of trichlorotriphenyl tetrazolium chloride (TTC), and the expression of NF- 魏 B in brain tissue was detected by immunohistochemistry. The content of TNF- 偽 in brain tissue was measured by enzyme linked immunosorbent assay (Elisa). Results the scores of Ludmila Belayev 12 in the model group were significantly higher than those in the normal group and the sham operation group, and the scores in the model group were significantly lower than those in the model group, and the volume of cerebral infarction in the model group was significantly larger than that in the normal group and the sham operation group (P 0.01), and compared with that in the model group, the cerebral infarction volume in the model group was significantly higher than that in the control group and the sham operation group. The positive expression rate of NF- 魏 B in the model group was significantly higher than that in the normal group and sham operation group. The expression of NF- 魏 B in the model group was significantly lower than that in the model group, and the content of TNF- 偽 in the model group was significantly higher than that in the model group. The content of TNF- 偽 in the normal group and the sham operation group was significantly lower than that in the model group. Conclusion PDTC can reduce the content of TNF- 偽 and decrease the expression of TNF- 偽 by inhibiting the activity of NF- 魏 B, so it may play a protective role in cerebral ischemia-reperfusion injury by regulating the expression of TNF- 偽 through NF- 魏 B, which may be related to the inhibition of inflammation.
