【摘要】：目的觀察腦卒中后抑郁(PSD)大鼠大腦情感障礙相關腦區(qū)額葉、海馬、杏仁核神經元數(shù)量變化。方法將20只健康成年雌性SD大鼠隨機分為抑郁組、卒中組、PSD組和對照組,每組5只。抑郁組采用孤養(yǎng)法加以慢性不可預見的中等強度應激刺激(CUMS)制作抑郁模型。卒中組用線栓法建立腦卒中模型。PSD組先用線栓法建立腦卒中模型,然后用孤養(yǎng)法加以CUMS制作PSD模型。對照組大鼠正常飼養(yǎng)。給予CUMS后第29天處死各組大鼠,取額葉、海馬、杏仁核區(qū)腦組織行神經元特異性核蛋白(NeuN)免疫熒光組織染色,光鏡下計數(shù)并比較各組大鼠額葉、海馬、杏仁核中神經元(NeuN陽性細胞)的數(shù)量。結果 PSD組大鼠情感障礙相關腦區(qū)額葉、海馬、杏仁核中神經元數(shù)量分別為(98.20±1.78)、(43.00±7.87)、(93.00±3.53)個/視野,均低于其他三組(P均0.05)。結論 PSD組大鼠情感障礙相關腦區(qū)額葉、海馬和杏仁核神經元數(shù)量減少。
[Abstract]:Objective to observe the changes of neurons in frontal lobe, hippocampus and amygdala in affective disorder related brain regions of rats with poststroke depression (PSD). Methods Twenty healthy adult female SD rats were randomly divided into depression group, stroke group and control group with 5 rats in each group. Depression model was established in depression group with chronic unpredictable moderate stress stimulation (CUMS). The stroke model was established by thread embolization method in stroke group. The stroke model was established by thread embolization method in PSD group, and then PSD model was made by CUMS. Rats in the control group were fed normally. On the 29th day after CUMS, the rats in each group were killed. The brain tissues of frontal lobe, hippocampus and amygdaloid area were stained with neuron-specific nucleoprotein Neun. The frontal lobe and hippocampus of each group were counted and compared under light microscope. The number of Neun positive neurons in the amygdala. Results the number of neurons in the frontal lobe, hippocampus and amygdala in the PSD group was 98.20 鹵1.78 and 93.00 鹵3.53 respectively, which was lower than that in the other three groups (P < 0.05). Conclusion the number of neurons in frontal lobe, hippocampus and amygdala decreased in PSD group.
