【摘要】：目的：探討ANRIL基因rs1333040、rs10757272和rs6475606三個(gè)單核苷酸多態(tài)(SNPs)與湖南漢族人群散發(fā)顱內(nèi)動(dòng)脈瘤(slA)的相關(guān)性。 方法：收集sIA患者108例及對(duì)照者107例的臨床資料及血液標(biāo)本,并提取其血液基因組DNA。應(yīng)用基質(zhì)輔助激光解吸電離飛行時(shí)間質(zhì)譜(MALDI-TOF-MS)技術(shù)對(duì)病例組和對(duì)照組ANRIL基因的rs1333040、rs10757272和rs6475606三個(gè)多態(tài)進(jìn)行基因分型檢測(cè)。采用多因素二元logistic回歸分析比較slA組與對(duì)照組、slA組中的單發(fā)組和多發(fā)組分別與對(duì)照組、單發(fā)slA組中的破裂組和未破裂組分別與對(duì)照組、單發(fā)slA組中的前群動(dòng)脈瘤組和后群動(dòng)脈瘤組分別與對(duì)照組該三個(gè)多態(tài)基因型及等位基因頻率的差異。 結(jié)果：(1)slA組和對(duì)照組ANRIL基因rs1333040、rs10757272、 rs6475606三個(gè)多態(tài)基因型及等位基因頻率比較均無(wú)統(tǒng)計(jì)學(xué)差異(p0.05,多因素二元logistic回歸)。(2)sIA組中的單發(fā)slA組和多發(fā)slA組ANRIL基因rs1333040、rs10757272、rs6475606三個(gè)多態(tài)基因型及等位基因頻率分別與對(duì)照組比較均無(wú)統(tǒng)計(jì)學(xué)差異(p0.05,多因素二元logistic回歸)。(3)單發(fā)slA組中的破裂sIA組和未破裂slA組ANRIL基因rs1333040、rs10757272、rs6475606三個(gè)多態(tài)基因型及等位基因頻率分別與對(duì)照組比較均無(wú)統(tǒng)計(jì)學(xué)差異(p0.05,多因素二元logistic回歸)。(4)單發(fā)slA組中的前群sIA組和后群slA組ANRIL基因rs1333040、rs10757272和rs6475606三個(gè)多態(tài)基因型及等位基因頻率分別與對(duì)照組比較均無(wú)統(tǒng)計(jì)學(xué)差異(p0.05,多因素二元logistic回歸)。 結(jié)論：(1)ANRIL基因rs1333040、rs10757272和rs6475606三個(gè)多態(tài)可能與湖南漢族人群sIA的易感不相關(guān)。(2)ANRIL基因rs1333040、rs10757272和rs6475606三個(gè)多態(tài)可能與湖南漢族人群slA單發(fā)/多發(fā),單發(fā)sIA破裂/未破裂患者發(fā)病風(fēng)險(xiǎn)不相關(guān)。(3)在湖南漢族單發(fā)sIA患者中,ANRIL基因rs1333040、rs10757272和rs6475606三個(gè)多態(tài)可能與顱內(nèi)動(dòng)脈瘤前/后群位置分布無(wú)關(guān)。
[Abstract]:Objective: to investigate the relationship between the ANRIL gene rs1333040rs10757272 and rs6475606 and the sporadic intracranial aneurysm SNPs in Hunan Han population. Methods: the clinical data and blood samples of 108 patients with sIA and 107 controls were collected and their blood genome was extracted. Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) was used to detect the polymorphism of ANRIL gene rs1333040 rs10757272 and rs6475606 in patients and controls. Multivariate logistic regression analysis was used to compare the single and multiple groups in slA group and control group, and the ruptured group and unruptured group in single slA group and control group respectively. The polymorphism genotype and allele frequency of anterior group aneurysm group and posterior group aneurysm group in single slA group were different from those in control group. Results the ANRIL gene rs1333040rs10757272 was not significantly different among the control group and the control group. There was no significant difference in the frequency of the three polymorphic genotypes and alleles among the three rs6475606 genotypes. There were three polymorphic genotypes of ANRIL gene rs1333040rs10757272rs6475606 in the multivariate logistic regression. There was no significant difference in allelic frequencies between the two groups (p0.05). There was no significant difference in allelic frequencies between the two groups (p0.05, multivariate binary logistic regression analysis) ANRIL gene rs1333040rs10757272rs6475606 in single slA group and unruptured slA group, respectively, as compared with the control group. There was no significant difference in ANRIL gene rs1333040rs10757272 and rs6475606 allele frequencies between pre- and posterior slA groups in multivariate logistic regression analysis and multivariate logistic regression analysis (P 0.05). There was no significant difference in ANRIL genotype and allelic frequencies between the two groups as compared with the control group. P0.05, multivariate logistic regression. Conclusion the three polymorphisms of the ANRIL gene rs1333040rs10757272 and rs6475606 may not be related to the susceptibility of sIA in Hunan Han population. The rs1333040 rs10757272 and rs6475606 polymorphisms of ANRIL gene may be singleton / multiplex with slA in Hunan Han population. The polymorphism of ANRIL gene rs1333040rs10757272 and rs6475606 may not be associated with the distribution of anterior / posterior group of intracranial aneurysms.
