恩再適對CCI大鼠痛覺過敏及脊髓NR2B、NF-κB和iNOS表達的影響

【摘要】:目的:1.觀察坐骨神經(jīng)慢性損傷大鼠機械痛閾及熱痛閾的變化,測定坐骨神經(jīng)慢性壓迫性損傷(Chronic constriction injury, CCI)大鼠脊髓N-甲基D-天冬氨酸受體2B亞基(N-methyl-D-aspartic acid receptor2B, NR2B)、核因子-κB(nuclear factor-kappa B, NF-κB)和誘導(dǎo)型一氧化氮合酶(Inducible nitric oxide synthase, iNOS)的表達,探討神經(jīng)病理性疼痛的發(fā)病機制。 2.觀察恩再適對CCI大鼠痛覺過敏及脊髓NR2B、NF-κB和iNOS表達的影響。 方法:1.56只180-220g雄性SD大鼠隨機分為2組:分別為Sham組(假手術(shù)組,n=8)和CCI組(CCI組,n=48)。于術(shù)前1d、術(shù)后1d、4d、7d、14d、21d測定機械痛閾(mechanical withdrawal threshold, MWT)和熱痛閾(paw withdrawal latency, PWL)后處死,取L4-6脊髓,采用逆轉(zhuǎn)錄-聚合酶鏈式反應(yīng)(Reverse transcription-Polymerase chain reaction, RT-PCR)和蛋白質(zhì)印跡法(Western blot, WB)檢測脊髓NR2B、NF-κB和iNOS的表達。 2.96只180-220g雄性SD大鼠隨機分為2組:分別CCI組(CCI模型組,n=48)和A組(恩再適封閉治療組,n=48)。測定MWT和PWL后處死大鼠,取L4-6脊髓,采用實時定量PCR(Real time quantitative PCR, qPCR)和WB方法檢測脊髓NR2B、NF-κB和iNOS的表達。 結(jié)果:1.與Sham組相比,CCI組術(shù)側(cè)從術(shù)后1d開始出現(xiàn)MWT值下降(P0.01,n=8),術(shù)后7d降至最低(P0.01,n=8)。術(shù)后14d時仍處于較低水平(P0.01,n=8)。與Sham組相比,CCI組術(shù)側(cè)從術(shù)后1d開始出現(xiàn)PWL值下降(P0.01,n=8),術(shù)后7d降至最低(P0.01,n=8)。術(shù)后14d時仍處于較低水平(P0.01,,n=8)。脊髓組織中NR2B和NF-κB于損傷后1d開始增加(P0.01,n=4);7d達到高峰,與術(shù)前相比有顯著性意義(P0.01,n=4)。術(shù)后14d雖有所降低,但仍維持在較高水平(P0.01,n=4)。iNOS mRNA的表達于術(shù)后1d開始增加(P0.01,n=4),術(shù)后14d達到高峰,顯著高于術(shù)前(P0.01,n=4),術(shù)后21d時仍保持較高水平表達(P0.01,n=4)。 iNOS mRNA與NR2BmRNA和NF-κB mRNA的表達呈顯著正相關(guān)(r=0.833,P0.05;r=0.842,P0.05)。 2.術(shù)后CCI組MWT及PWL較術(shù)前明顯降低(n=8,P0.01),A組MWT及PWL明顯高于CCI組(n=8, P0.05)。 CCI組術(shù)后脊髓NR2B、NF-κB和iNOS的表達較術(shù)前明顯升高(n=4,P0.05),A組脊髓NR2B、NF-κB和iNOS的表達較CCI組明顯降低(n=4,P0.05)。 結(jié)論:1.CCI術(shù)后大鼠脊髓中NR2B、NF-κB及下游iNOS表達增加,可能與神經(jīng)病理性疼痛的產(chǎn)生和維持有關(guān)。 2.恩再適封閉治療能夠改善CCI大鼠痛覺過敏,其作用效應(yīng)與影響脊髓NR2B、NF-κB和iNOS的表達有關(guān)。
[Abstract]:Purpose 1.To observe the changes of mechanical pain threshold and thermal pain threshold in rats with chronic sciatic nerve injury.The expression of N-methyl-D-aspartic acid receptor 2B subunit N-methyl-D-aspartic acid receptor 2B, nuclear factor- 魏 B(nuclear factor-kappa B (NF- 魏 B) and inducible nitric oxide synthase (iNOS) in the spinal cord of rats with chronic compression injury of sciatic nerve was determined.To explore the pathogenesis of neuropathic pain.2.To observe the effects of enrezol on hyperalgesia and the expression of NF- 魏 B and iNOS in spinal cord of CCI rats.Methods 1.56 male Sprague-Dawley rats (180-220 g) were randomly divided into two groups: Sham group (sham-operated group) and CCI group.The mechanical withdrawal threshold (MWT) and the thermal pain threshold of paw withdrawal (PWL) were measured 1 day before operation and 4 days after operation. The spinal cord L4-6 was killed. Reverse transcription-Polymerase chain reactionation (RT-PCR) and Western blotting were used to detect the expression of NR2BN-NF- 魏 B and iNOS in spinal cord.2.96 male Sprague-Dawley rats (180-220 g) were randomly divided into two groups: CCI group (CCI model group) and group A (group A).MWT and PWL were used to determine the expression of NF- 魏 B and iNOS in spinal cord NR2BN- 魏 B, and the L4-6 spinal cord was harvested. PCR(Real time quantitative PCR (qPCR) and WB were used to detect the expression of NF- 魏 B and iNOS in spinal cord.The result is 1: 1.Compared with the Sham group, the MWT value decreased from the first day after operation to the lowest value at the 7th day after operation.On the 14th day after operation, it was still at a lower level (P 0.01).Compared with the Sham group, the PWL value decreased from the first day after operation to the lowest value at the 7th day after operation.On the 14th day after operation, it was still at a lower level (P 0.01).The levels of NR2B and NF- 魏 B in spinal cord tissue began to increase at 1 day after injury and reached the peak at 7 days after injury, which was significantly higher than that before operation.On the 14th day after operation, the expression of P0. 01 mRNA was maintained at a high level, but the expression of iNOS mRNA increased on the 1st day after operation, and reached the peak on the 14th day after operation.The expression of iNOS mRNA was significantly higher than that of preoperation (P 0.01), and remained high at 21 days after operation. There was a significant positive correlation between the expression of iNOS mRNA and NR2BmRNA and NF- 魏 B mRNA.2.The MWT and PWL in CCI group were significantly lower than those in pre-operation group (P 0.01). The MWT and PWL in group A were significantly higher than those in group CCI (P 0.05).The expression of NF- 魏 B and iNOS in spinal cord of CCI group was significantly higher than that before operation. The expression of NF- 魏 B and iNOS in group A was significantly lower than that in group CCI.Conclusion 1. The increased expression of NF- 魏 B and downstream iNOS in spinal cord of rats after CCI may be related to the generation and maintenance of neuropathic pain.2.Enreclosure block therapy can improve hyperalgesia in CCI rats, and its effect is related to the effect on the expression of NF- 魏 B and iNOS in spinal cord NR2BU.
