細胞周期依賴激酶抑制劑Olomoucine對活化星形膠質(zhì)細胞神經(jīng)蛋白聚糖、短蛋白聚糖表達的抑制作用

【摘要】:目的研究細胞周期依賴激酶抑制劑Olomoucine對星形膠質(zhì)細胞活化后神經(jīng)蛋白聚糖(Neurocan)、短蛋白聚糖(Brevican)表達的抑制作用。方法應(yīng)用睫狀神經(jīng)營養(yǎng)因子(CNTF)刺激法建立體外培養(yǎng)星形膠質(zhì)細胞的活化模型,實驗分3組:對照組、活化組與抑制組。對照組:正常培養(yǎng)的星形膠質(zhì)細胞;活化組:星形膠質(zhì)細胞加入20 ng/ml睫狀神經(jīng)營養(yǎng)因子(CNTF)處理24 h;抑制組:星形膠質(zhì)細胞用20 ng/ml睫狀神經(jīng)營養(yǎng)因子預(yù)處理24 h,加入100μmol/L Olomoucine。應(yīng)用ELISA法檢測不同時間點(12 h、24 h、48 h)各組細胞上清液中Neurocan、Brevican含量變化,半定量RT-PCR檢測各組細胞GFAP mRNA及Neurocanm RNA、Brevican mRNA的表達變化。結(jié)果(1)活化組上清液中Neurocan、Brevican的含量在12 h、24 h、48 h隨著時間的延長逐漸增加,與對照組比較差異有顯著性(P0.01);抑制組上清液中Neurocan、Brevican含量隨著時間的延長較活化組明顯降低,與對照組、活化組比較差異有顯著性(P0.01)。(2)活化組細胞GFAP mRNA、Neurocan mRNA、Brevican mRNA表達明顯增加,與對照組比較差異有顯著性(P0.01);抑制組細胞GFAP mRNA、Neurocan mRNA、Brevican mRNA表達均明顯下調(diào),與活化組比較差異有顯著性(P0.01)。結(jié)論細胞周期依賴激酶抑制劑Olomoucine可抑制活化星形膠質(zhì)細胞神經(jīng)蛋白聚糖、短蛋白聚糖表達。
[Abstract]:Objective to study the inhibitory effect of cell cycle dependent kinase inhibitor (Olomoucine) on the expression of neuroproteoglycan neurocanine (Brevican) after astrocyte activation. Methods the cultured star was established by ciliary neurotrophic factor (ciliary neurotrophic factor) stimulation in vitro. The activation model of glial cells, The experiment was divided into three groups: control group, activated group and inhibitory group. Control group: normal cultured astrocytes; In activated group, astrocytes were treated with 20 ng/ml ciliary neurotrophic factor (CNTF) for 24 h, while in inhibitory group, astrocytes were pretreated with 20 ng/ml ciliary neurotrophic factor for 24 h and added 100 渭 mol/L Olomoucine. ELISA assay was used to detect 12 h and 24 h after treatment. The content of Neurocanium Brevican in the supernatant of cells in each group was changed for 48 h. Semi-quantitative RT-PCR was used to detect the expression of GFAP mRNA and Neurocanm RNA-Brevican mRNA. Results the content of Neurocann Brevican in supernatant of activated group increased gradually with the prolongation of time at 12 h, 24 h and 48 h, respectively. Compared with the control group, the content of Neurocann Brevican in the supernatant of the inhibitory group was significantly lower than that in the activated group, and the expression of GFAP mRNA-NeuromRNA-Brevican mRNA in the activated group was significantly higher than that in the control group. Compared with the control group, the expression of GFAP mRNA-neuromRNA-Brevican mRNA was significantly down-regulated in the inhibition group, and the difference was significant compared with the activated group. Conclusion Olomoucine, a cell cycle dependent kinase inhibitor, can inhibit the activation of astrocyte neuroproteoglycan. Short proteoglycan expression.
