顳葉癲癇模型幼鼠和成年大鼠發(fā)作時間、發(fā)作類型的差異及發(fā)作后Ang-2蛋白表達變化

【摘要】:目的對比觀察顳葉癲癇模型幼鼠和成年大鼠在發(fā)作時間、發(fā)作類型的差異,分析癲癇發(fā)作是否具有年齡依賴性;并觀察兩組大鼠顳葉癲癇發(fā)作后海馬區(qū)血管內(nèi)皮系統(tǒng)促血管生成素2(Ang-2)的蛋白表達變化及差異。方法選取健康雄性Wistar大鼠,分為幼年組(2~3周,體重30~60 g)和成年組(7~8周,體重200~250 g),各32只,均隨機分為正常對照組(12只)及實驗組(20只)。按Racine分級標準觀察大鼠行為改變,并記錄各組大鼠癲癇發(fā)作時間及情況。正常對照組腹腔注射生理鹽水。正常對照組大鼠癲癇持續(xù)狀態(tài)后第1、3、5、7天頸椎脫臼處死,斷頭取海馬組織,采用Western blot技術(shù)觀察Ang-2的蛋白表達變化。結(jié)果正常對照組大鼠均未發(fā)生任何癲癇樣發(fā)作表現(xiàn);實驗組大鼠從給藥至出現(xiàn)癲癇樣發(fā)作Ⅳ級時間,幼鼠為(12.7±5.12)min,成年鼠為(25.3±6.71)min;幼鼠癲癇發(fā)作程度較成年鼠嚴重;首發(fā)癲癇持續(xù)狀態(tài)(SE)達Ⅳ級及Ⅳ級以上者占90%,成年鼠為50%;癲癇發(fā)作后10%成年鼠可自行終止,幼鼠不能自行終止,且均發(fā)展為癲癇持續(xù)狀態(tài);幼鼠死亡率10%,成年鼠無死亡。幼鼠及成年大鼠的正常對照組在腹腔注射生理鹽水后第1、3、5、7天,海馬中Ang-2蛋白表達無明顯改變(P0.01);與正常對照組相比,癲癇持續(xù)狀態(tài)后幼鼠及成年大鼠海馬組織Ang-2蛋白表達均升高(P0.05),且幼鼠升高程度較成年鼠高(P0.05)。癲癇持續(xù)狀態(tài)后第1天起,幼鼠及成年鼠海馬中Ang-2蛋白表達均逐漸升高,第5天達高峰,后下降(P0.05)。結(jié)論癲癇發(fā)作具有年齡依賴性;Ang-2蛋白表達水平與癲癇發(fā)作相關(guān)。
[Abstract]:Objective to compare the time and type of temporal lobe epilepsy between young and adult rats, and to analyze whether the seizure is age-dependent. The changes and differences of protein expression of vascular endothelial system angiopoietin-2 (Ang-2) in hippocampal area after temporal lobe epilepsy were observed in two groups. Methods healthy male Wistar rats were divided into juvenile group (3 weeks) and adult group (30 ~ 60 g) and adult group (7 ~ 8 weeks). A total of 32 rats were randomly divided into normal control group (n = 12) and experimental group (n = 20). The behavioral changes of rats were observed according to Racine grading standard. The time and condition of epileptic seizures in each group were recorded. The normal control group was injected with normal saline intraperitoneally. The rats in the normal control group were killed by cervical dislocated and severed hippocampus on the 7th day after epileptic status. Western blot technique was used to observe the expression of Ang-2 protein. Results there was no epileptic seizure in the normal control group, while in the experimental group, the time from the administration of the drug to the onset of epileptiform seizure grade 鈪,
