【摘要】：目的觀察Sirt1對(duì)神經(jīng)母細(xì)胞瘤SK-N-SH細(xì)胞系中Tau蛋白磷酸化的影響。方法在體外培養(yǎng)的SK-N-SH細(xì)胞中加入腺病毒過(guò)表達(dá)的Sirt1和SirtM(Sirt mutant),利用Western blot檢測(cè)細(xì)胞中Sirt1和SirtM的表達(dá),并通過(guò)Western blot、Real-time PCR、免疫細(xì)胞化學(xué)技術(shù)檢測(cè)t-Tau蛋白以及p-Tau蛋白的表達(dá),流式細(xì)胞術(shù)(FCM)檢測(cè)Sirt1對(duì)SK-N-SH細(xì)胞凋亡的影響。結(jié)果 Western blot結(jié)果顯示,Sirt1組及SirtM組相比于對(duì)照組t-Tau蛋白的水平降低,Sirt1組較SirtM組相比降低較為顯著;Tau蛋白的磷酸化位點(diǎn)ser404、thr231更為明顯;Real-time PCR結(jié)果顯示,Sirt1組相比于對(duì)照組Tau的mRNA水平下降;免疫細(xì)胞化學(xué)結(jié)果顯示,Sirt1可以降低Tau蛋白及其磷酸化的水平;流式細(xì)胞術(shù)檢測(cè)提示Sirt1組較對(duì)照組降低了SK-N-SH細(xì)胞的凋亡。結(jié)論Sirt1能夠降低神經(jīng)母細(xì)胞瘤SK-N-SH中Tau蛋白的表達(dá)及磷酸化、減少細(xì)胞的凋亡;本研究為T(mén)au蛋白病等神經(jīng)退行性疾病的研究提供了重要的實(shí)驗(yàn)依據(jù)。
[Abstract]:Objective to observe the effect of Sirt1 on the phosphorylation of Tau protein in SK-N-SH cell line of neuroblastoma. Methods Adenovirus-overexpression Sirt1 and SirtM(Sirt mutant were added to SK-N-SH cells in vitro. The expression of Sirt1 and SirtM was detected by Western blot. The expression of t-Tau protein and p-Tau protein were detected by Western blotReal-time PCR and immunocytochemistry. Results the effect of Sirt1 on apoptosis of SK-N-SH cells was detected by flow cytometry. Results the results of Western blot showed that the level of t-Tau protein in Western blot group and SirtM group was lower than that in control group. The phosphorylation site ser404ht231 of tt1 group was significantly lower than that of SirtM group. The results of real-time PCR showed that the mRNA level of Tau in Sirt1 group was lower than that in control group. The results of immunocytochemistry showed that Tau protein and phosphorylation could be reduced by sirt1. Flow cytometry showed that the apoptosis of SK-N-SH cells in Sirt1 group was lower than that in control group. Conclusion Sirt1 can reduce the expression and phosphorylation of Tau protein and apoptosis in SK-N-SH of neuroblastoma. This study provides an important experimental basis for the study of neurodegenerative diseases such as Tau's disease.
